      Tingkat Stres Ayam IPB-D1 dan IPB-D2 Berdasarkan Rasio Heterofil/Limfosit Dalam Darah

      Abstract
      Ayam IPB-D1 dan IPB-D2 dikembangkan secara khusus dari hasil silangan ayam Cobb, pelung, sentul, dan kampung dengan harapan dihasilkannya ras ayam yang unggul dan memiliki kekebalan yang lebih tinggi terhadap penyakit jika dibandingkan dengan tetuanya. Ketahanan terhadap penyakit dipengaruhi oleh berbagai faktor salah satunya adalah hormon yang bisa menjadi indikator stres. Penelitian ini dilakukan untuk mengetahui tingkat stres ayam IPB-D1 dan IPB-D2 berdasarkan rasio H/L (heterofil/limfosit) dalam darah, sehingga daya tahan tubuh ayam yang diteliti dapat teramati. Sebanyak masing-masing 25 dan 30 ekor ayam IPB-D1 dan IPB-D2 digunakan dalam penelitian ini. Preparat ulas darah dibuat dengan pewarnaan Giemsa dari darah yang diambil dari v. brachialis. Sediaan diamati dengan mikroskop perbesaran 100x10. Jenis leukosit diidentifikasi dengan melihat literatur sebagai acuan dan dihitung menggunakan aplikasi Haematology Counter hingga mencapai 100 buah, kemudian dihitung rasio H/L-nya. Hasil penelitian menunjukkan rasio H/L ayam IPB-D2 lebih rendah dari ayam IPB-D1, dan lebih rendah dari para tetuanya. Dengan suhu lingkungan di luar suhu ideal ayam broiler, ayam IPB-D2 dapat beradaptasi dengan baik tanpa menimbulkan efek stres fisiologis jika dibandingkan dengan ayam IPB-D1.
       
      IPB-D1 and IPB-D2 chickens were one of local breeds that were developed by crossbreeding of Cobb parent stock, Pelung, Sentul, and Kampoong chickens. The advantage of this chickens is their resistance against Newcastle Diseases virus infection but the stress level of this chicken has not yet been studied. This study aimed to determine the stress levels of IPB-D1 and IPB-D2 chickens by analyzing the H/L (heterophile/lymphocyte) ratio in their blood, in order to observe the chickens' resistance. A total of 25 IPB-D1 chickens and 30 IPB-D2 chickens were used for the research. Blood smears were taken from v. brachialis and stained with Giemsa. Using a 100×10 magnifying microscope, the blood smears were examined, leukocyte types were identified, and the H/L ratio was calculated. The results showed that the H/L ratio of IPB-D2 chickens was lower than that of IPB-D1 chickens and their origin breeds. This shows that IPB-D2 chickens, as opposed to IPB-D1 chickens, have greater ability to adapt to environmental temperatures outside of the ideal range for broiler chickens without showing any signs of physiological stress.
