      Praperlakuan Peracetic Acid Pada Ekstraksi Ulvan Dari Ulva lactuca Dengan Akselerasi Ultrasonikasi

      Abstract
      Ulvan merupakan heteropolisakarida bersulfat diekstrak dari dinding sel rumput laut Ulva yang memiliki aktivitas biologis potensial untuk pangan, biomedik, pertanian. Hasil ekstraksi ulvan kerap kali mempunyai rendemen rendah dan berwarna gelap kehitaman. Penambahan kecepatan pengadukan dan metode praperlakuan pemutihan peracetic acid (PAA) dipilih sebagai metode alternatif dalam ekstraksi ulvan dengan ultrasonik untuk mendapatkan hasil yang efisien. Peracetic acid adalah oksidator kuat yang digunakan sebagai pemutih. Penelitian ini bertujuan menentukan pengaruh ultrasonikasi, suhu, pengadukan serta praperlakuan pemutihan terhadap rendemen ulvan dan karakteristiknya. Ulvan dari rumput laut Ulva lactuca diberi prapelakuan dengan PAA kemudian diekstraksi dengan pelarut akuades pada suhu 40, 60, 80°C selama 90 menit dengan berbantu ultrasonikasi. Suhu secara signifikan meningkatkan nilai rendemen. Praperlakuan bleaching dangan PAA dan ultrasonikasi secara signifikan dapat meningkatkan rendemen ulvan. Pengadukan tidak berpengaruh secara signifikan meningkatkan nilai rendemen. Perlakuan ulvan terpilih yaitu pretreatment bleaching, suhu 80°C, kecepatan pengadukan 400 rpm yang menghasilkan rendemen lebih banyak, warna yang cerah, bobot molekul rendah.       Ulvan is a sulfated heteropolysaccharide extracted from the cell wall of Ulva seaweed which has potential biological activity for food, biomedical, agriculture. Ulvan extraction often results low yield and dark color. Stirring speed and pretreatment peracetic acid method was chosen as an alternative method in ultrasonic extraction to obtain efficient results. Peracetic acid (PAA) is a strong oxidizing agent used as a bleach. This study aims to determine the effect of ultrasonication, temperature, stirring and bleaching pretreatment on the yield and characteristics of ulvan. Ulvan was pretreated using PAA and then extracted from Ulva lactuca using distilled water at 40, 60, 80°C for 90 minutes with the aid of ultrasonication. PAA pretreatment and ultrasonication could significantly increase of ulvan yield. Furthermore, temperature significantly increases the yield value. Stirring has no significant effect on increasing the yield value. The selected ulvan treatment was bleaching pretreatment, temperature 80°C, stirring speed 400 rpm which produced more yield, bright color, low molecular weight.
