      Karakteristik Molekuler Gen COI pada Amfibi (Ordo: Anura) sebagai Dasar Pengelolaan di Lingkungan Kampus IPB Dramaga

      Abstract
      Amfibi merupakan vertebrata yang memiliki peran penting sebagai bioindikator lingkungan. Anura (katak dan kodok) merupakan ordo amfibi yang membutuhkan ketersediaan air dalam siklus hidupnya. Identifikasi molekuler dapat digunakan untuk memantau keberadaan Anura di habitat alami melalui data informasi genetik. Penanda gen cytochrome oxidase subunit I (COI) direkomendasikan sebagai gen yang efektif untuk mengidentifikasi amfibi menggunakan DNA barcoding. Penelitian ini bertujuan mengidentifikasi karakteristik gen COI pada amfibi (Ordo: Anura) di lingkungan kampus IPB. Penelitian ini dilaksanakan pada bulan Agustus sampai Desember 2022 di kampus IPB Dramaga. Data penelitian berasal dari hasil sekuensing (17 sekuen spesimen katak dan kodok) yang belum diolah dan dianalisis sebelumnya. Analisis validasi spesies katak dan kodok pada panjang fragmen 647-786 bp, menunjukkan tingkat kemiripan berkisar 82,74-100%. Spesies katak dan kodok ordo Anura memiliki hubungan kekerabatan yang dekat dan nilai jarak genetik sebesar 0,000-0,259. Terdapat 31 situs nukleotida spesifik sebagai pembeda dari spesies katak dan kodok ordo Anura. Nilai keragaman haplotype dan keragaman nukleotida dari 3 lokasi menunjukkan keragaman genetik yan tinggi.
       
      Amphibians are vertebrates that have an important role as environmental bioindicators. Anura (frogs and toads) is an amphibian order that requires water availability in its life cycle. Molecular identification can be used to monitor the presence of Anura in natural habitats through genetic information data. The cytochrome oxidase subunit I (COI) gene marker is recommended as an effective gene for identifying amphibians using DNA barcoding. This study aims to identify the characteristics of the COI gene in amphibians (Order: Anura) in the IPB campus environment. This research was conducted from August to December 2022 at IPB Dramaga campus. The research data came from sequencing results (17 sequences of frog and toad specimens) that had not been processed and analyzed before. Validation analysis of frog and toad species at fragment lengths of 647-786 bp, showing similarity levels in the range of 82.74-100%. The frog and toad species of the Anura order have a close relationship and the genetic distance value is 0.000-0.259. There are 31 specific nucleotide sites to distinguish frog and toad species of the Anura order. The value of haplotype diversity and nucleotide diversity from 3 locations shows high genetic diversity.
