      Gambaran Hematologi dan Aktivitas Enzimatis Hati Tikus Hiperkolesterolemia yang Diberikan "Emping-Dadih"

      Abstract
      Hypercholesterolemia is a condition of high cholesterol in the blood. It can lead to death due to occurring cardiovascular disease. Prevention and treatment of hypercholesterolemia using traditional food black glutinous rice and fermented buffalo milk (“emping-dadih or ED”) may affect blood profiles and liver enzymatic activity. This study aimed to analyze various parameters of red blood cells, white blood cells, and thrombocytes, also liver enzymatic activities of rat giving high-cholesterol and ED. This study was conducted using 25 male Sprague-Dawley rats divided into five treatment groups, namely negative control (A), positive control/hypercholesterolemia (B), ED prevention (C), ED treatment (D), and atorvastatin treatment (E). Hematology and liver enzymatic comparison for all groups were analyzed by descriptive and ANOVA. The results showed that ED increased the number of erythrocytes, decreased the number of platelets and leukocytes as indicators decreased of inflammation. “Emping-dadih” also significantly reduced SGPT/ALT and SGOT/AST activities than in hypercholesterolemia conditions. This study showed that “emping-dadih” was able to improve the hematological profile and enzymatic activity of the liver in hypercholesterolemia rats.
       
      Hiperkolesterolemia adalah suatu kondisi kolesterol yang tinggi di dalam darah. Hal ini dapat menyebabkan kematian karena memicu terjadinya penyakit kardiovaskular. Pencegahan dan pengobatan hiperkolesterolemia menggunakan pangan tradisional beras ketan hitam dan dadih (“emping-dadih” atau ED) dapat memengaruhi profil darah dan aktivitas enzimatis hati. Penelitian ini bertujuan menganalisis berbagai parameter sel darah merah, sel darah putih, dan trombosit, serta aktivitas enzimatis hati tikus yang diberikan kolesterol dan ED. Penelitian ini dilakukan dengan menggunakan 25 ekor tikus jantan Sprague-Dawley yang dibagi menjadi lima kelompok perlakuan, yaitu kontrol negatif (A), kontrol positif/hiperkolesterolemia (B), pencegahan ED (C), pengobatan ED (D), dan pengobatan atorvastatin (E). Perbandingan hematologi dan enzimatis hati untuk semua kelompok dianalisis secara deskriptif dan ANOVA. Hasil penelitian menunjukkan bahwa ED meningkatkan jumlah eritrosit, menurunkan jumlah trombosit dan leukosit sebagai indikator penurunan inflamasi. “Emping-dadih” juga secara signifikan mengurangi aktivitas SGPT/ALT dan SGOT/AST dibandingkan dengan kondisi hiperkolesterolemia. Penelitian ini menunjukkan bahwa “emping-dadih” mampu memperbaiki profil hematologi dan aktivitas enzimatis hati pada tikus hiperkolesterolemia.
