      Pengaruh Minyak Cengkeh, Gliserol, Pembekuan, dan Suhu Pembugaran terhadap Survival Rate Ikan Kapiat (Barbonymus schwanenfeldii)

      Abstract
      Teknologi transportasi ikan hias hidup yang sesuai tuntutan komoditas sangat diperlukan. Penelitian ini bertujuan untuk menentukan konsentrasi anestesi minyak cengkeh, pengaruh gliserol, lama waktu pembekuan, serta suhu media pembugaran terbaik sehingga menghasilkan nilai survival rate ikan kapiat (Barbonymus schwanenfeldii) yang tinggi serta mengamati perubahan histologi insang. Penelitian ini mengaplikasikan anestesi minyak cengkeh, perendaman dalam larutan gliserol 5%, pembekuan bersuhu -13ºC dalam rentang waktu 10, 15, dan 20 menit, serta pembugaran pada media bersuhu ruang (28oC) dan 10oC. Konsentrasi optimum anestesi minyak cengkeh adalah 25 ppm dengan rata-rata waktu menjelang pingsan 3,70±0,20 menit dan menjelang bugar 2,57±0,39 menit. Durasi pembekuan terbaik adalah 10 menit, sedangkan suhu media pemulihan terbaik adalah 28°C dengan perolehan survival rate sebesar 100%. Perendaman dalam larutan gliserol 5% belum efektif untuk melindungi jaringan insang akibat pembekuan. Perubahan histologi berupa pembendungan darah, pendarahan, dan lepasnya lamela sekunder.
       
      The technology for transporting live ornamental fish that meets the demands of the commodity is highly necessary. This research aims to determine the concentration of clove oil anesthesia, the effect of glycerol, the duration of freezing, and the best recovery media temperature in order to produce a high survival rate of kapiat fish (Barbonymus schwanenfeldii) and observe changes in gill histology. This research applied clove oil anesthesia, immersing in a 5% glycerol solution, freezing at a temperature of -13ºC for a duration of 10, 15, and 20 minutes, and recovery in media with room temperature (28oC) dan 10oC. The optimum concentration of clove oil anesthesia is 25 ppm with an average time of unconsciousness of 3,70±0,20 minutes and recovery time of 2,57±0,39 minutes. The best freezing duration is 10 minutes, while the best recovery media temperature is 28°C with a survival rate of 100%. Immersing in a 5% glycerol solution is not yet effective in protecting the gill tissue from freezing. Histological changes include blood congestion, bleeding, and detachment of secondary lamellae.
