      Isolasi dan seleksi kapang nematofagus untuk pengendalian haemonchosis domba

      Abstract
      Dari 200 sampel tanah yang telah dikumpulkan dari beberapa tempat di daerah Bogor (Ciapus, Darmaga, Kebun Raya Bogor, Lab Terpadu FKH IPB /Laladu dan Taman Kencana) dilakukan isolasi kapang nematofagus dengan metode Janssons (1985) dan Larsen et al. (1991). Isolat yang ditemukan diperbanyak dengan metode sel spora tunggal yang kemudian diindentifikasi menurut Barron (1977) dan Gronvold et al. (1996). Setelah diperbanyak, diseleksi dengan uji simulasi saluran pencernaan ruminansia (ludah sintetik, cairan rumen, larutan pepsin dan tripsin) dengan metode Larsen et al. (1991). Setelah melalui uji ini, calon kapang nematofagus diuji kemampuan keefektifannya dalam membuat jerat serta kemampuan efisiensi membunuh larva infektif menurut metode Nansen et al. (1988). Hasil isolasi diperoleh kapang Arthrobotrys oligospora dan Duddingtonia flagrans isolat dalam negeri yang kemudian dibandingkan keefektifitannya dengan isolat luar negeri. Hasil akhir seleksi penelitian ini menunjukkan bahwa A. oligospora isolat dalam negeri kurang keefektifannya dalam membuat jerat dan efisiensi membunuh larva infektif bila dibandingkan dengan isolat A. oligospora luar negeri.
       
      A total of 200 soil samples collected from some locations Bogor of areas (Botanical Garden, Ciapus, Darmaga, Kencana Park, Laladu), were used to isolate nematophagous fungi from soil using Janssons (1985) and Larsen et al. Method (1991). The isolates were recovered by single cell spore technique and identified by Barron (1977) and Gronvold et al. (1996). They were then multiplied and selected in vitro stress by modified Larsen et al. Method (1991), initiating the condition of ruminant stomach (rumen fluid, pepsin and tripsin) and synthetic saliva. Through this test, the viable fungi were then tested their capability in making traps and trapping efficiency against Haemonchus contortus larvae, by Nansen et al. method (1988). The result indicated that local isolate of Arthrobotrys oligospora was recovered as candidate of nematophagous fungi from Indonesia. This isolate was then compared to A. oligospora and Duddingtonia flagrans from overseas. It was resulted by testing that A. oligospora from overseas was better than local A. oligospora and D. flagrans in trap making capability and trapping efficiency.
