      Abstract
      Pada proses maturasi in vitro oosit distimulasi untuk mencapai pertumbuhan yang sempurna hingga mencapai tahap metafase II (MII). Oosit yang secara morfologi telah terseleksi dengan baik berdasarkan jumlah dan kekompakan sel kumulus serta sitoplasma yang homogen, masih berbeda-beda dalam kualitas dan kemampuan perkembangannya. Hal ini memungkinkan oosit yang dimaturasi mencapai tahap MII tidak bersamaan. Oosit yang sudah mencapai tahap MII harus segera difertilisasi karena apabila tidak difertilisasi selama waktu optimal fertilisasi, oosit akan mengalami aging. Terjadinya aging pada oosit tahap MII disebabkan karena penurunan aktivitas maturation promoting factor (MPF). Kafein melalui mekanismenya dapat menghambat aktivitas Wee1 dan Myt1 sehingga terjadi perubahan dari pre-MPF menjadi MPF aktif, hal ini mengakibatkan tingginya aktivitas MPF.
       
      In the process of in vitro maturation, oocytes were stimulated to achieve an optimum growth to reach metaphase II (MII) stage. Oocytes morphologically which properly selected based on the number and compactness of the cumulus cells and homogeneous cytoplasm are still vary in quality and developmental competence. This allows oocytes matured reaching MII stage did not uniform. Oocytes which have reached MII stage should be fertilized immeadiately because if not fertilized during the optimal time of fertilization, oocyte will be aging. The occurrence of aging on MII oocytes caused by a decrease in maturation promoting factor (MPF) activity. Trough the mechanism of caffeine can inhibit the activity of Wee1 and Myt1 resulting in a change of pre-MPF become active MPF, this results in a high MPF activity.
