Status Fitonematoda pada Tanaman Jahe di Wilayah Bogor, Cianjur, dan Sukabumi, Jawa Barat

Infeksi nematoda parasit tumbuhan atau fitonematoda pada pertanaman jahe menjadi ancaman serius dalam budi daya jahe karena dapat menurunkan kuantitas dan kualitas rimpang jahe. Rimpang jahe terinfeksi fitonematoda dapat menjadi media penyebarannya di lapangan dan berdampak pada penerimaan pasar domestik maupun internasional terutama untuk ekspor rimpang segar. Dilaporkan adanya penolakan ekspor rimpang jahe segar Indonesia ke Vietnam dan Jepang karena terinfeksi nematoda parasit tumbuhan.
Informasi tentang kelimpahan fitonematoda dan jenis nematoda pada tanaman jahe serta gambaran petani tentang pengelolaannya sangat penting. Hal itu berkontribusi dalam pengelolaan kesehatan dan penekanan penyakit tanaman oleh petani. Penelitian ini bertujuan untuk mengetahui kelimpahan populasi dan mengidentifikasi fitonematoda penting pada pertanaman jahe serta mengetahui beberapa pengendalian yang dilakukan oleh petani di wilayah Bogor, Cianjur, dan Sukabumi, Jawa Barat.
Penelitian ini dilakukan dalam empat tahap, yaitu: (1) pengambilan sampel rimpang, akar, dan tanah dari pertanaman jahe di wilayah Bogor, Cianjur, dan Sukabumi. (2) ekstraksi dan analisis komunitas fitonematoda. (3) identifikasi fitonematoda (4) wawancara dengan petani jahe di wilayah Bogor, Cianjur, dan Sukabumi. Tahap pertama dilakukan pengambilan sampel rimpang, akar, dan tanah dari pertanaman jahe yang menunjukkan gejala infeksi fitonematoda yang berasal dari wilayah produksi jahe Bogor, Cianjur, dan Sukabumi, Jawa Barat.
Tahap kedua adalah ekstraksi sampel rimpang, akar, dan tanah dan analisis komunitas fitonematoda. Fitonematoda dari sampel rimpang dan akar diekstraksi dengan menggunakan metode mist chamber yang dimodifikasi. Fitonematoda dari sampel tanah diekstraksi menggunakan metode flotasi sentrifugasi. Analisis komunitas fitonematoda dengan peubah yang diamati yaitu, populasi absolut, frekuensi absolut, dan nilai prominensi untuk mengetahui fitonematoda yang dominan pada pertanaman jahe.
Tahap ketiga adalah identifikasi fitonematoda berdasarkan karakter morfologi, morfometri dan molekuler. Identifikasi karakter morfologi dan morfometri dimulai dengan membuat slide permanen dari fitonematoda yang telah difiksasi, kemudian diamati menggunakan mikroskop cahaya. Fitonematoda diamati dari bagian ujung bibir (anterior) hingga bagian ujung ekor (posterior). Identifikasi karakter molekuler dilakukan pada spesies Meloidogyne sp. untuk mengkonfirmasi karakter morfologi dan morfometri. Identifikasi molekuler Meloidogyne sp. dilakukan dengan teknik Polymerase Chain Reaction (PCR).
Pengumpulan data petani melalui metode wawancara dengan panduan kuesioner terstruktur. Petani responden berasal dari tiga wilayah, yaitu Bogor, Cianjur, dan Sukabumi. Responden di masing-masing wilayah sebanyak 20 orang ditentukan secara sengaja, yaitu petani jahe yang memiliki pengalaman dalam budi daya jahe minimal dalam waktu satu tahun terakhir. Tabulasi data dengan menggunakan Microsoft Excel 2016 dan dianalisis secara deskriptif.
Petani dari lokasi pengambilan sampel, menanam jenis jahe merah varietas unggul Jahira 2. Bibit jahe yang digunakan petani berasal dari hasil panen musim tanam sebelumnya. Petani di wilayah Bogor menerapkan pola tanam monokultur dan sebagian besarnya menanam jahe dalam polybag. Sementara petani di wilayah Cianjur dan Sukabumi menerapkan pola tanam polikultur dengan tanaman cabai, jagung, timun, terong, peria, dan kacang panjang. Petani melakukan pengolahan tanah dan pemberian pupuk organik sebelum tanam. Mayoritas petani melakukan pengolahan tanah minimum.
Gejala penyakit tanaman yang disebabkan oleh fitonematoda di lapangan umumnya mengelompok dan tidak merata. Gejala tanaman yang terinfeksi fitonematoda di permukaan tanah berupa daun menguning, tanaman layu, jumlah anakan berkurang, dan tanaman kerdil. Tanda serangan fitonematoda yang ditemukan pada rimpang berupa lesi atau luka berbentuk cekungan basah, dan pada rimpang yang diiris ditemukan saluran infeksi dengan warna coklat gelap. Tanda infeksi fitonematoda terlihat pada jumlah akar yang berkurang dan bekas luka dengan warna coklat kemerahan pada jaringan akar, yang disebabkan oleh fitonematoda yang bersifat endoparasit berpindah. Selain itu juga ditemukan juga pembengkakan pada bagian akar atau puru akar yang disebabkan oleh fitonematoda yang bersifat endoparasit menetap
Hasil identifikasi karakter morfologi ditemukan sebanyak tujuh spesies fitonematoda yang berasosiasi dengan tanaman jahe di wilayah Bogor, Cianjur, dan Sukabumi. Berdasarkan cara parasitismenya pada tanaman diklasifikasikan menjadi 4 kategori. Kategori pertama terdiri dari fitonematoda ektoparasit, yaitu Aphelenchoides bicaudatus, Hoplolaimus sp., dan Helicotylenchus sp. Kategori kedua adalah endoparasit berpindah antara lain Pratylenchus sp. dan Radopholus similis. Kategori ketiga adalah semi-endoparasit yaitu Rotylenchulus reniformis, dan kategori terakhir adalah endoparasit menetap yaitu Meloidogyne sp.
Hasil analisis komunitas fitonematoda ditemukan dua spesies yang dominan, yaitu nematoda Radopholus similis yang dominan ditemukan pada jahe asal Bogor dan nematoda Meloidogyne sp. dominan ditemukan pada tanaman jahe asal Sukabumi dan Cianjur. Identifikasi molekuler dilakukan pada fitonematoda Meloidogyne sp. yang memiliki panjang amplikon 170 pb pada daerah amplifikasi gen Cytochrome c oxidase subunit 1 (CoxI) dan didapatkan spesies Meloidogyne javanica dengan tingkat homologi 98,2 % terhadap isolat pembanding di GenBank.
Hasil wawancara dengan petani responden menunjukkan bahwa banyak dari mereka memiliki pemahaman yang baik tentang cara penyebaran penyakit, karakteristik bibit yang sehat, dan jenis hama dan penyakit pada tanaman jahe. Namun, kebanyakan petani masih belum mengetahui istilah Pengendalian Hama Terpadu (PHT), perbedaan gejala yang disebabkan oleh fitonematoda dan penyakit lainnya, serta peran musuh alami. Penerapan pengendalian terpadu yang telah dilakukan petani meliputi penggunaan pestisida alami dan kimia, penggunaan varietas unggul, pengamatan lapang, dan perlakuan bibit sebelum tanam. Metode pengendalian yang masih belum diterapkan secara optimal oleh petani adalah sanitasi dan rotasi tanam. The infection of plant-parasitic nematodes, also known as phytonematodes, in ginger farming has become a major threat to the cultivation of ginger. The infection can result in a decrease in both the quantity and quality of the ginger rhizomes. Moreover, the infected rhizomes can act as carriers, facilitating the spread of the nematodes in the fields and could negatively affect the acceptance of the domestic and international markets, particularly when exporting fresh rhizomes. It has been reported that exports of Indonesian fresh ginger rhizomes to Vietnam and Japan were rejected due to the presence of phytonematodes.
Information about the abundance of phytonematodes and identification of nematode species in ginger plants, as well as the farmer's perspective on integrated management, contributes to the plant's health management and disease suppression by farmers and related industries. This study aims to determine the population abundance, identify important phytonematodes in ginger farming, and understand the control measures taken by farmers in Bogor, Cianjur, and Sukabumi, West Java.
The study was conducted in four stages: (1) the sampling of rhizome, root, and soil from ginger farms in the areas of Bogor, Cianjur, and Sukabumi, (2) the extraction and analysis of the phytonematodes community, (3) the identification of phytonematodes, (4) the interviews with ginger farmers in Bogor, Cianjur, and Sukabumi. In the first stage, rhizome, root, and soil samples were taken from ginger farms showing symptoms of phytonematodes infection from Bogor, Cianjur, and Sukabumi, West Java.
The second stage was extracting rhizome, root, and soil samples and analysis of the phytonematodes community. The phytonematodes from rhizome and root samples were extracted using a modified mist chamber method, while those from soil samples were extracted using the flotation centrifugation method. The phytonematodes community was analyzed based on the observed variables of absolute population, absolute frequency, and prominence value to determine the dominant phytonematodes in ginger farming.
The third stage was the identification of phytonematodes based on morphological, morphometric, and molecular characters. The morphological and morphometric identification of phytonematodes began by making a permanent slide of the fixed phytonematodes, which were then observed using a light microscope. Phytonematodes were observed from the anterior end to the posterior end. The molecular identification of Meloidogyne sp. was carried out to confirm the morphological and morphometric characteristics. The molecular identification of Meloidogyne sp. was conducted using Polymerase Chain Reaction (PCR) technique.
Data from farmers was collected through structured questionnaire interviews. The respondents were from three areas: Bogor, Cianjur, and Sukabumi. The 20 respondents from each area were determined randomly, consisting of farmers with at least one year of experience in ginger cultivation. Data tabulation was done using Microsoft Excel 2016 and analyzed descriptively.
The farmers from the sample collection location planted red ginger with the superior variety Jahira 2. The farmer used ginger seed from the previous planting season's harvest. Farmers in the Bogor region apply a monoculture planting pattern and most plant ginger in polybags. Meanwhile, farmers in the Cianjur and Sukabumi regions apply a polyculture planting pattern with chili, corn, cucumber, eggplant, shallots, and long beans. Farmers performed soil preparation and organic fertilization before planting, with most farmers performing minimum soil preparation.
The symptoms of plant diseases caused by phytonematodes are not evenly distributed and are grouped in the field. Symptoms of plants infected with phytonematodes on the soil surface include yellowing leaves, wilting, decreased number of shoots, and stunted growth. On the rhizomes, signs of phytonematode attack include lesions or moist depression-shaped wounds and dark brown infectious channels when sliced. Root tissue may show reduced numbers of roots and scar-shaped wounds with a reddish-brown color caused by endoparasitic migratory. Additionally, swelling of the root or the presence of root galls can also indicate endoparasitic sedentary.
The morphological character identification results revealed seven phytonematodes associated with ginger plants in Bogor, Cianjur, and Sukabumi. The phytonematodes were categorized based on their mode of parasitism on the plant. The first category consisted of ectoparasitic phytonematodes, including Aphelenchoides bicaudatus, Hoplolaimus sp., and Helicotylenchus sp. The second category was migratory endoparasites, represented by Pratylenchus sp. and Radopholus similis. The third category was semi-endoparasitic, represented by Rotylenchulus reniformis, and the fourth and final category was sedentary endoparasitic, represented by Meloidogyne sp.
The result of phytonematode community analysis found two dominant species, Radopholus similis which was dominant in ginger from Bogor, and Meloidogyne sp., which was dominant in ginger from Sukabumi and Cianjur. Molecular identification was performed on Meloidogyne sp. phytonematodes with a 172 bp amplicon length in the Cytochrome c oxidase subunit 1 (CoxI) amplification region, and the species Meloidogyne javanica was obtained with a 98.2% homology level compared to reference isolates in GenBank.
The interview results with the respondent farmers showed that many have a good understanding of how the disease spreads, the characteristics of healthy seedlings, and the types of pests and diseases in ginger plants. However, most farmers still need to learn about the term Integrated Pest Management (IPM), the difference in symptoms caused by phytonematodes and other diseases, and the role of natural enemies. The common methods of control employed by farmers include using natural and chemical pesticides, superior plant varieties, monitoring the fields, and treating seedlings before planting. Additional measures require optimization in their application, such as implementing sanitary measures, rotating crops, and utilizing natural enemies.
