Metabolomik Rempah Minor Indonesia famili Zingiberaceae dan Potensinya sebagai Antibakteri

Penelitian ini bertujuan untuk (1) melakukan penapisan aktivitas antibakteri dari 12 spesies rempah minor Zingiberaceae dengan uji difusi agar sumur (2) menentukan satu rempah terpilih untuk dilanjutkan ke tahapan fraksinasi dengan aktivitas antibakteri tertinggi, (3) mengorelasikan data profil kimia HPLC berupa selang waktu retensi dan aktivitas antibakteri dengan OPLS untuk memperoleh selang waktu retensi yang paling berkorelasi dengan aktivitas antibakteri dari fraksi teraktif dan (4) mengidentifikasi komponen aktif yang berpotensi sebagai antibakteri dari selang waktu retensi terpilih hasil analisis OPLS dengan melakukan analisis UHPLC-HRMS fraksi teraktif dimana waktu retensi tersebut dominan. Hasil penapisan 12 rempah Zingiberaceae menunjukkan bahwa ekstrak Curcuma purpurascens Blume paling efektif menghambat bakteri S. aureus dibanding rempah lainnya. Curcuma purpurascens Blume juga menunjukkan zona hambat yang cukup besar pada bakteri E. coli meski bukan yang tertinggi. Selanjutnya Curcuma purpurascens Blume dilanjutkan ke tahapan berikutnya (identifikasi komponen aktif). Hasil pengukuran rata-rata diameter zona hambat fraksi Curcuma purpurascens Blume menunjukkan bahwa fraksi metanol memiliki aktivitas antibakteri tertinggi dengan rata-rata diameter zona hambat sebesar 18,12±2,34 mm terhadap S. aureus. Identifikasi senyawa aktif yang berpotensi sebagai antibakteri menggunakan pendekatan metabolomik dengan cara mengorelasikan nilai aktivitas antibakteri dengan profil kimia berupa selang waktu retensi kromatogram dari HPLC menggunakan analisis data multivariat OPLS. Berdasarkan nilai koefisien korelasi dan variabel of influence (VIP), selang waktu retensi yang berkorelasi dengan aktivitas antibakteri adalah selang waktu retensi 16,5-18,0 menit. Identifikasi senyawa aktif yang berpotensi sebagai antibakteri dilakukan dengan cara mencocokkan pola kromatogram fraksi metanol dari HPLC dengan pola kromatogram fraksi metanol dari UHPLC-HRMS. Puncak pada waktu retensi 17,8 menit terdeteksi pada selang waktu retensi 16,5-18,0 menit pada kromatogram HPLC. Puncak ini berkorelasi dengan puncak waktu retensi 16,5 menit pada kromatogram UHPLC-HRMS. Berdasarkan pola fragmentasi spektra massa yang dicocokkan dengan pangkalan data PubChem senyawa yang berhasil diidentifikasi pada waktu retensi 16,5 menit pada UHPLC-HRMS adalah senyawa demethoxycurcumin. This study aims to (1) screen the antibacterial activity of 12 Zingiberaceae minor herb species by agar well diffusion test, (2) determine which herb is selected to proceed to the fractionation stage with the highest antibacterial activity, (3) correlate HPLC chemical profile data in the form of time intervals retention and antibacterial activity with OPLS to obtain the retention time interval that is most correlated with the antibacterial activity of the most active fraction and (4) identify active components that have potential as antibacterial from selected retention intervals resulting from OPLS analysis by conducting UHPLC-HRMS analysis of the most active fraction where retention time is dominant. The screening results of 12 Zingiberaceae spices showed that Curcuma purpurascens Blume extract was the most effective against S. aureus bacteria compared to other spices. Curcuma purpurascens Blume also showed a fairly large inhibition zone for E. coli bacteria, although not the highest. Furthermore, Curcuma purpurascens Blume continued to the fractionation stage. The results of measuring the average inhibition zone diameter of the Curcuma purpurascens Blume fraction showed that the methanol fraction had the highest antibacterial activity with an average inhibition zone diameter of 18.12 ± 2.34 mm against S. aureus. Identifying active compounds that have potential as antibacterial used a metabolomics approach by correlating antibacterial activity values with chemical profiles in the form of HPLC retention time intervals through OPLS analysis. OPLS results showed that the retention time interval that correlates with antibacterial activity is a retention time interval of 16.5-18.0 minutes. Identification of active compounds that have potential as antibacterial were done by matching the chromatogram pattern of the methanol fraction from HPLC with the chromatogram pattern of the methanol fraction from UHPLC-HRMS. The peak at a retention time of 17.8 minutes was detected at a retention time interval of 16.5-18.0 minutes on the HPLC chromatogram. This peak associated with a retention time of 16.5 minutes obtained from UHPLC-HRMS. Based on fragmentation pattern comparison with the PubChem library, the compound identified at a retention time of 16.5 minutes on the UHPLC-HRMS was demethoxycurcumin.
