Studi Koinfeksi Kecacingan dan Parasit Darah pada Kerbau Lumpur di Kabupaten Sumba Timur

Kabupaten Sumba Timur dikenal sebagai salah satu lumbung ternak di wilayah timur Indonesia. Kerbau lumpur adalah salah satu komoditas ternak yang banyak dikembangbiakkan oleh masyarakat setempat. Kerbau lumpur memiliki banyak peran dalam sistem sosial ekonomi masyarakat setempat. Pemanfaatannya tidak hanya berkaitan dengan pekerjaan sehari-hari, namun juga berperan dalam berbagai macam upacara adat. Beberapa tahun terakhir dilaporkan terjadinya penurunan populasi kerbau di Kabupaten Sumba Timur. Penurunan ini diduga berkaitan dengan adanya infeksi penyakit, salah satunya adalah infeksi parasit. Infeksi parasit yang terjadi umumnya ditemukan baik dalam bentuk infeksi tunggal dan koinfeksi. Penelitian mengenai interaksi koinfeksi antara beberapa parasit pada ternak di Indonesia sampai saat ini belum banyak dilaporkan, meskipun secara empiris hampir semua hewan terinfeksi lebih dari satu jenis parasit (multiparasitisme). Koinfeksi parasit kemungkinan dapat menjadi faktor penyebab kematian ternak yang tinggi jika dilihat dari efek yang ditimbulkannya pada inang. Penelitian mengenai interaksi koinfeksi parasit pada ternak di Indonesia, khususnya di Kabupaten Sumba Timur, penting dilakukan mengingat dampak koinfeksi yang dapat ditimbulkan. Penelitian yang dilakukan bertujuan untuk mengidentifikasi jenis parasit, membandingkan kelompok infeksi tunggal dan koinfeksi terhadap kondisi kesehatan kerbau, serta menganalisis hubungan antara intensitas infeksi kecacingan terhadap beberapa variabel yang berkaitan dengan tingkat keparahan infeksi pada kerbau lumpur di Kabupaten Sumba Timur yang mengalami koinfeksi.
Penelitian ini merupakan penelitian fundamental dengan pendekatan eksploratif. Sebanyak 105 contoh tinja dan ulas darah yang berasal dari kerbau lumpur di Kabupaten Sumba Timur diperiksa untuk mengetahui jenis parasit yang menginfeksi. Pemeriksaan contoh tinja menggunakan metode flotasi sederhana, modifikasi McMaster, dan filtrasi bertingkat. Contoh ulas darah diperiksa dengan mengamati dan memeriksa keberadaan parasit darah secara langsung di bawah mikroskop. Faktor yang memengaruhi tingkat keparahan infeksi juga diperiksa, seperti nilai hitung beda leukosit dan konsentrasi total protein. Nilai hitung beda leukosit dilakukan dengan menghitung secara relatif masing-masing jenis leukosit. Konsentrasi total protein ditentukan dengan menggunakan metode Bradford pada contoh serum kerbau yang ada. Seluruh data yang diperoleh ditabulasikan, disajikan secara deskriptif, dan dianalisis secara statistik untuk mengetahui signifikansi perbedaan hasil. Analisis statistik dengan uji korelasi Pearson juga dilakukan untuk menganalisis hubungan intensitas kecacingan dengan beberapa variabel yang memengaruhi tingkat keparahan infeksi.
Terdapat beberapa jenis parasit yang menginfeksi kerbau lumpur di Kabupaten Sumba Timur. Parasit cacing yang ditemukan meliputi kelompok Nematoda gastrointestinal (GI), Cestoda, dan Trematoda, sedangkan protozoa meliputi kelompok Apicomplexa GI dan darah. Kerbau lumpur di Kabupaten Sumba Timur diketahui mengalami koinfeksi dengan berbagai kelompok parasit (62,86%) dibandingkan dengan infeksi tunggal (37,14%). Terdapat setidaknya tiga bentuk
koinfeksi yang ditemukan dalam studi ini, yaitu koinfeksi cacing dan parasit darah, protozoa GI dan parasit darah, serta koinfeksi ketiganya. Koinfeksi cacing dan parasit darah memiliki nilai persentase infeksi tertinggi (27,62%) di antara seluruh bentuk koinfeksi, namun tidak ditemukan adanya bentuk koinfeksi antara cacing dan protozoa GI. Penelitian ini juga tidak menemukan pola infeksi tunggal kelompok cacing pada kerbau. Pola infeksi tunggal hanya disebabkan oleh parasit darah (36,19%), dengan nilai persentase infeksi antara kerbau yang mengalami infeksi tunggal dan koinfeksi tidak berbeda signifikan. Hasil studi menunjukkan bahwa sebanyak 104 ekor kerbau (99,05%) menderita infeksi parasit darah. Koinfeksi merupakan pola infeksi utama pada kerbau lumpur di Sumba Timur, dengan persentase infeksi sebesar 64,76% yang didominasi oleh koinfeksi Babesia sp. dengan Anaplasma sp. (47,62%). Infeksi tunggal parasit darah ditemukan 34,29%. Sebanyak 26,67% infeksi tunggal tersebut disebabkan oleh Anaplasma sp. Kerbau jantan (73,33%) diketahui lebih rentan terinfeksi oleh parasit cacing dan protozoa GI dibandingkan dengan kerbau betina (60,00%). Hasil serupa juga ditunjukkan oleh persentase infeksi parasit darah.
Intensitas infeksi parasit pada kerbau lumpur di Kabupaten Sumba Timur secara umum dapat dikategorikan ringan. Kerbau lumpur di Kabupaten Sumba Timur memiliki nilai intensitas infeksi kecacingan dan parasit darah yang beragam. Hasil studi menunjukkan bahwa intensitas infeksi tertinggi disebabkan oleh infeksi cacing Moniezia benedeni (547,03 telur tiap gram tinja/TTGT), diikuti oleh infeksi protozoa GI Eimeria sp. (220,69 oosista tiap gram tinja/OTGT), dan cacing Toxocara vitulorum (194,11 TTGT). Intensitas infeksi parasit darah tertinggi disebabkan oleh infeksi Trypanosoma evansi (0,50%), sedangkan intensitas infeksi terendah (0,15%) disebabkan oleh Theileria sp. Perbedaan nilai intensitas infeksi juga diperlihatkan pada pola infeksi yang berbeda. Kelompok kerbau yang mengalami infeksi tunggal parasit darah diketahui memiliki nilai intensitas infeksi yang lebih tinggi (0,33 ± 0,04) dibandingkan dengan kelompok koinfeksi (0,22 ± 0,08). Analisis statistik juga menunjukkan perbedaan yang signifkan (p<0,05) terhadap kelompok infeksi tunggal dengan kelompok koinfeksi.
Analisis hubungan terhadap intensitas infeksi kecacingan terhadap beberapa faktor yang memengaruhi keparahan infeksi (variabel uji) dilakukan dengan menentukan nilai koefisien korelasi Pearson. Hasil analisis menunjukkan bahwa terdapat variasi kekuatan dan arah hubungan intensitas infeksi kecacingan dengan variabel uji. Pada studi ini didapatkan bahwa penentuan korelasi intensitas infeksi kecacingan terhadap intensitas infeksi T. evansi tidak dapat diukur karena tidak terpenuhinya syarat uji. Intensitas infeksi kecacingan memiliki korelasi sangat lemah terhadap umur, nilai hitung beda leukosit, dan intensitas infeksi Babesia sp., dan Anaplasma sp. Korelasi dengan kategori cukup ditemukan pada nilai intensitas infeksi Theileria sp. Hampir seluruh variabel menunjukkan korelasi yang tidak searah, kecuali pada nilai monosit, neutrofil, dan intensitas infeksi Babesia sp. dan Theileria sp. Analisis statistik yang dilakukan menunjukkan bahwa seluruh hasil analisis korelasi tidak memiliki nilai yang signifikan (p>0,05). East Sumba Regency is one of the livestock production areas in eastern Indonesia. Swamp buffalo is one of the livestock commodities widely bred and has many roles in the socio-economic system of the local community. Its use is not only related to a source of food and labor but also plays a role in various traditional ceremonies. In recent years, it has been reported that there has been a decline in the buffalo population in East Sumba Regency. This decrease is thought to be related to infectious diseases, one of which is a parasitic infection. Parasitic infections that occur are generally found in both single and coinfection forms. Research on coinfection interactions between several parasites in livestock in Indonesia has not been widely reported. However, almost all animals are infected with more than one type of parasites (multiparasitism). Parasitic coinfection is likely to be a factor in high livestock mortality when viewed from the effect it has on the host. Research on the interaction of parasite coinfection in livestock in Indonesia, especially in East Sumba Regency, is important considering the impact of coinfection that can be caused. The research aimed to identify the type of parasite, compare groups of single infection and coinfection to the health condition of buffaloes, and analyze the relationship between the intensity of helminth infection and several variables related to the severity of infection in swamp buffaloes in East Sumba Regency who were coinfected.
This research is fundamental research with an exploratory approach. A total of 105 samples of feces and blood smears from swamp buffalo in East Sumba Regency were examined to determine the type of parasite that infected. The stool samples were examined using a simple flotation method, modified McMaster, and multilevel filtration. Blood samples are examined by observing and examining the presence of blood parasites directly under a microscope. Factors affecting the severity of infection were also examined, such as the differential leukocyte count and total protein concentration. The value of the different leukocyte counts is done by calculating the relative value of each type of leukocyte. The total protein concentration was determined using the Bradford method on buffalo serum samples. All data obtained were tabulated, presented descriptively, and statistically analyzed to determine the significance of differences in results. Statistical analysis using the Pearson correlation test was also carried out to analyze the relationship between helminth intensity and several variables that affect the severity of infection.
Several types of parasites infect swamp buffalo in East Sumba Regency. The helminth parasites found included the gastrointestinal (GI) Nematodes, Cestodes, and Trematodes, while the protozoa included the Apicomplexa GI and hemoprotozoa (blood parasites). Swamp buffaloes in East Sumba Regency were coinfected with various groups of parasites (62.86%) compared to a single infection (37.14%). There are at least three forms of coinfection found in this study, namely coinfection of helminths and blood parasites, GI protozoa, and blood parasites, and coinfection of all three. Coinfection of helminths and blood parasites had the highest percentage of infection (27.62%) among all forms of coinfection, but no
form of coinfection between helminths and GI protozoa was found. This study also did not find a single infection pattern of worms in buffaloes. The pattern of single infection was only caused by blood parasites (36.19%), with the percentage value of infection between buffaloes experiencing single infection and coinfection not significantly different. The results of the study showed that as many as 104 buffalo (99.05%) suffered from blood parasite infections. Coinfection was the main infection pattern in swamp buffalo in East Sumba, with a percentage of infection of 64.76%, which was dominated by Babesia sp. with Anaplasma sp. (47.62%). Blood parasite single infection was found in 34.29%. Anaplasma sp. caused a total of 26.67% of these single infections. Male buffalo (73.33%) are more susceptible to infection by GI worms and protozoa than female buffalo (60.00%). The percentage of blood parasite infections also showed similar results.
In general, the intensity of parasitic infection in swamp buffalo in the East Sumba Regency can be categorized as mild. Swamp buffaloes in East Sumba Regency have various intensity values of helminth infections and blood parasites. The results of the study showed that the highest infection intensity was caused by infection with Moniezia benedeni (547.03 eggs per gram of feces/EPG), followed by infection with the GI protozoan Eimeria sp. (220.69 oocysts per gram of feces/OPG), and Toxocara vitulorum (194,11 EPG). The highest infection intensity of blood parasites was caused by Trypanosoma evansi infection (0.50%), while the lowest infection intensity (0.15%) was caused by Theileria sp. Differences in infection intensity values are also shown in different infection patterns. The buffalo group with a single blood parasite infection had a higher infection intensity value (0.33 ± 0.04) compared to the coinfected group (0.22 ± 0.08). Statistical analysis also showed a significant difference (p<0.05) between the single infection group and the coinfected group.
Analysis of the relationship between the intensity of helminth infections and several factors that influence the severity of infection (test variables) was carried out by determining the Pearson correlation coefficient. The statistical analysis showed variations in the strength and direction of the relationship between the intensity of helminth infections and the test variables. This study found that the correlation between the intensity of helminth infection and the intensity of Trypanosoma evansi infection could not be measured because the test requirements were not fulfilled. The intensity of helminth infection has a very weak correlation with age, different leukocyte count values, and the intensity of infection of Babesia sp. and Anaplasma sp. Correlation with sufficient category was found in the intensity value of Theileria sp. Almost all variables showed a non-unidirectional correlation, except for the value of monocytes, neutrophils, and the intensity of Babesia sp. and Theileria sp. Statistical analysis showed that all the results of the correlation analysis did not have a significant value (p>0.05).
