      Karakteristik Protein Serisin Ulat Sutra Bombyx mori L. pada Ras dan Konsentrasi Pelarut Berbeda

      Abstract
      Protein serisin merupakan produk samping dari industri sutra yang memiliki struktur biokimia dan sifat biofisik yang unik. Penelitian ini bertujuan mengidentifikasi karakteristik protein serisin yang terdapat pada kokon ras ulat sutra Bombyx mori L. dengan konsentrasi pelarut degumming (NaOH) yang berbeda. Penelitian menggunakan rancangan acak kelompok faktorial dengan faktor perbedaan ras ulat sutra dan konsentrasi pelarut degumming (NaOH). Analisis yang dilakukan yaitu analisis rendemen serisin, analisis bobot molekul serisin dan analisis asam amino. Proses ekstraksi serisin menghasilkan rendemen protein serisin sebesar 2,6%±0,5 pada konsentrasi NaOH 0% dan 29,8%±6,57 pada konsentrasi 33,3% NaOH 0,1 N. Hasil penelitian menunjukan bahwa penambahan pelarut NaOH pada proses degumming berpengaruh nyata (P<0,05) pada rendemen serisin yang dihasilkan, sedangkan perbedaan ras tidak berpengaruh nyata pada rendemen serisin (P>0,05). Bobot molekul serisin yang dihasilkan berkisar 15,68- 87,87 kDa. Asam amino protein terlarut serisin dikarakterisasi menggunakan high performance liquid chromatography (HPLC) dan dihasilkan asam amino tertinggi pada glisina (30,87%).
       
      Sericin protein is a by-product of the silk industry which has a unique biochemical structure and biophysical properties silkworm races Bombyx mori L. with different concentrations of degumming solvent (NaOH). The study used a factorial randomized block design with different factors of silkworm race and concentrations of degumming solvent degumming (NaOH). The analyzes carried out were sericin yield analysis, sericin molecular weight analysis and amino acid analysis. The sericin extraction process resulted in sericin protein yield of 2,6%±0,5 at 0% NaOH concentration and 29,8%±6,57 at 33,3% NaOH 0,1 N concentration. The results showed that the addition of NaOH solvent in the degumming had a significant effect (P<0,05) on the yield of sericin produced, while racial differences had no significant effect on the yield of sericin (P>0,05). The molecular weight of sericin produced ranged from 15,68 to 87,87 kDa. The soluble protein amino acid sericin was characterized using high performance liquid chromatography (HPLC) and the highest amino acid yield was glycine (30,87%).
