Struktur Genetika Populasi Ikan Tongkol Kawakawa Euthynnus affinis (Cantor 1849) di Wilayah Perairan Pulau Sumatera

Ikan tongkol kawakawa (KAW) (Euthynnus affinis) (Cantor, 1849) hidup secara berpindah-pindah pada zona epipelagis-neritik. Spesies ini tersebar luas di perairan tropis dan subtropis terbuka (oseanik) atau dekat dengan garis pantai (neritik) dengan kisaran suhu perairan 18-29℃. Kawakawa diketahui memiliki karakteristik epipelagic migratory yang melakukan migrasi atau berenang dengan jarak jauh untuk mencari makan dan memijah. Kawakawa masuk ke dalam kelompok tuna kecil dan memiliki ciri khusus dibandingkan ikan tongkol lainnya yaitu terdapat garis-garis hitam miring melengkung pada bagian atas gurat sisi dan memiliki titik hitam sejumlah 1-4 buah dekat sirip pektoral. Ikan ini bersifat predator opoturnistik dalam mencari makan, selain itu juga memakan cumi, krustasea, moluska, dan zooplankton. Penelitian ini bertujuan untuk mengkaji pola struktur keragaman genetika populasi kawakawa dan filogenetik dari beberapa populasi di wilayah perairan Pulau Sumatera; Belawan, Padang, dan Lampung dengan menggunakan DNA Mitokondria bagian kontrol (control region) d-loop.
Sebanyak 77 sampel ikan kawakawa berhasil diidentifikasi pada tingkat kecocokan 97-100% dari pusat data NCBI (National Center for Biotechnology Information). Tingkat keragaman genetika (h) dari ketiga lokasi penelitian relatif tinggi dengan nilai keragaman 0.9477-0.9900. Struktur genetik dari hasil analisis menunjukkan bahwa tidak terdapat perubahan struktur genetik yang dimiliki yang signifikan. P-value menunjukkan nilai 0.29423, dapat dikatakan bahwa tidak ada perbedaan antar populasi. Analisis distribusi haplotipe memperlihatkan adanya keterkaitan pada tiap haplotipe ikan kawakawa. Konektivitas antar haplotipe ikan kawakawa paling besar adalah pada haplotipe 4 terdiri dari 12 sampel yang menunjukkan adanya pencampuran individu dari ketiga populasi penelitian. Haplotipe 4 tersusun atas 5 individu dari populasi Belawan (42.66 %), 4 individu dari populasi Lampung (33.33 %) dan 3 individu dari populasi Padang (25 %). Sedangkan pada haplotipe 3 terdiri dari 6 sampel dengan pencampuran 4 individu dari populasi Belawan dan masing-masing 1 individu dari populasi Lampung dan Padang. Haplotipe 1 hanya terdapat pada populasi Belawan. Analisis keragaman genetik dan strukutur populasi ikan kawakawa di wilayah perairan Pulau Sumatera tidak membentuk struktur populasi. Komposisi haplotipe yang terdeteksi pada populasi penelitian mengindikasikan terdapat konektivitas dan kekerabatan genetik. The kawakawa tuna (KAW) (Euthynnus affinis) (Cantor, 1849) lives a nomadic life in the epipelagic-neritic zone. This species is widely distributed in open tropical and subtropical waters (oceanic) or close to the shoreline (neritic) with a water temperature range of 18-29℃. Kawakawa is known to have epipelagic migratory characteristics that migrate or swim long distances to find food and spawn. Kawakawa belongs to the small tuna group and has special characteristics compared to other tuna fish, such as black oblique and curved lines on the top of the lateral line also have 1-4 black dots near the pectoral fins. This fish is an opportunistic predator in search of food; eats squid, crustaceans, molluscs, and zooplankton. This study aims to examine the pattern of genetic diversity of the Kawakawa population and the phylogenetic structure of several populations in the waters of Sumatra Island: Belawan, Padang, and Lampung using mitochondrial DNA in the control region (control region) d-loop.
77 kawakawa muscle tissue samples were obtained from three fish landing ports and 77 samples were identified as Euthynnus affinis at a match rate of 97-100% from the NCBI (National Center for Biotechnology Information) data center. The level of genetic diversity (h) in the three populations is relatively high with a diversity value of 0.9477-0.9900. Genetic structure analysis shows that there is no significant difference in genetics. P-value shows a value of 0.29423, it can be said that there is no distinction between populations because the value is significant. The haplotype distribution analysis indicates that there was a relationship between each haplotype of kawakawa. The greatest connectivity between haplotypes of kawakawa in Hap-4 mixing of 5 individuals from the Belawan population (42.66%), 4 individuals from the Lampung population (33.33%), and 3 individuals from the Padang population (25%). Meanwhile, hap-3 consisted of 6 samples with a mixture of 4 individuals from the Belawan population and 1 individual each from the Lampung and Padang populations. Hap-1 is only found in the Belawan population. Analysis of genetic diversity and population structure of kawakawa in the Sumatra water does not form a population structure. Haplotype materials found in the study population found genetic connectivity and kinship.
