      Abstract
      Ikan kembung lelaki (Rastrelliger kanagurta) memiliki nilai ekonomis penting bagi nelayan. Ikan pelagis kecil di WPPNRI 572 dan WPPNRI 573 telah mengalami kondisi fully-exploited. Tekanan eksploitasi yang tinggi akan menurunkan keragaman genetik spesies. Penelitian ini bertujuan untuk memastikan spesies, menganalisis kekerabatan dan keragaman genetik secara molekuler ikan kembung lelaki dengan menggunakan marka gen COI di Perairan Selat Sunda, Cilacap dan Banyuwangi. Isolasi dan ekstraksi DNA menghasilkan 30 DNA total, lalu diamplifikasi dengan metode PCR. Hasil sekuensing divalidasi dengan fitur BLASTn pada aplikasi MEGA 11, menunjukkan bahwa 30 contoh dari ke tiga lokasi merupakan spesies R. kanagurta dengan kemiripan antara 98,15% hingga 100%. Jarak genetik ikan kembung lelaki di tiga lokasi memiliki nilai yang rendah. Hasil pohon filogeni menunjukkan contoh memiliki hubungan kekerabatan yang dekat dan adanya pencampuran populasi dari tiga lokasi, sehingga dapat dikelola dalam satu unit pengelolaan.
       
      The Indian Mackerel has important economic value for fishermen. The small pelagic fisheries in WPPNRI 572 and WPPNRI 573 have experienced a fully-exploited condition. High exploitation pressure will reduce the genetic diversity of Indian Mackerel species in the wild. This study aims to determine the species, analyze the molecular kinship and genetic diversity of Indian Mackerel (Rastrelliger kanagurta) using COI gene markers in Sunda Strait waters, Cilacap and Banyuwangi. DNA isolation and extraction yielded 30 total DNA, amplified by the PCR method. Sequencing results were validated with the BLASTn feature on the MEGA 11 application, showing that 30 samples were R. kanagurta species with the highest similarity of 100%. The genetic distance of Indian Mackerel in three locations has a low value. The results of the phylogeny tree show that the sample has a close relationship, and there is a mixing of populations from three locations so that it can be managed in one management unit.
