Marigold merupakan salah satu komoditas hortikultura tanaman hias yang banyak ditanam di Indonesia. Masyarakat mengenal tanaman ini karena aroma yang menyengat, dikenal dengan nama bunga “tahi kotok” atau “telek-telekan” dalam Bahasa Jawa dan bunga “gumitir” dalam Bahasa Bali. Keragaman tanaman Tagetes sp. masih tergolong rendah, oleh karena itu, untuk meningkatkan keragaman tanaman marigold dilakukan mutasi induksi kimia dengan EMS (Ethyl Methane Sulfonate). Pada penelitian ini, penulis melakukan aplikasi mutagen kimia EMS secara akut dan kronik pada benih Tagetes sp. Dalam metode aplikasi akut, mutagen kimia diberikan satu kali, dengan taraf konsentrasi tinggi sehingga kemampuan tanaman untuk bertahan hidup, serta banyaknya mutan yang dihasilkan semakin rendah. Sebagai alternatif untuk meningkatkan diperolehnya mutan yang berhasil hidup, metode aplikasi mutagen EMS secara kronik dilakukan dengan memberikan taraf konsentrasi EMS yang rendah dengan mengencerkan konsentrasi EMS pada LC50 dan merendam benih dalam jangka waktu yang lebih lama.
Penelitian ini bertujuan untuk mendapatkan nilai LC50 (Lethal Concentration 50), mengetahui tingkat sensitivitas tanaman, dan mendapatkan keragaman tanaman Tagetes sp. dengan mutagen EMS melalui metode aplikasi akut dan kronik. Materi genetik yang digunakan dalam penelitian ini adalah benih T. erecta genotipe MG04 dan T. patula genotipe MG21. Teknik mutasi akut dilakukan dengan merendam benih dalam larutan EMS selama 4 jam dengan 10 taraf konsentrasi, yaitu 0%, 0,2%; 0,4%; 0,6%; 0,8%; 1,0%; 1,5%; 2,0%; dan 3,0%. Teknik mutasi kronik dilakukan dengan mengencerkan konsentrasi EMS LC50 1/10x, 1/100x, 1/1000x dan lama perendaman 6 jam, 24 jam dan 48 jam. Pengamatan dilakukan terhadap parameter persentase tanaman hidup, karakter kuantitatif dan karakter kualitatif. Hasil penelitian menunjukkan nilai LC50 T. erecta sebesar 0,82% dan LC50 T. patula sebesar 1,87% yang berarti tingkat sensitivitas T. erecta terhadap perlakuan EMS lebih tinggi dibandingkan T. patula. Metode aplikasi akut menunjukkan tingkat mutasi yang relatif rendah dan keragaman bentuk tipe bunga terbatas. Metode aplikasi kronik menghasilkan bentuk bunga beragam dan warna bunga lebih putih pada T. erecta dan T. patula menghasilkan warna bunga lebih merah dibandingkan metode aplikasi akut.
Metode aplikasi akut T. erecta generasi M2 menghasilkan 4 tanaman mutan putatif warna putih yaitu M2-0,2%-20-1; M2-0,4%-39-1; M2-0,6%-20-2; M2-0,8%-20-2. Ada 2 mutan putatif warna kuning yaitu M2-1,0%-36-2; M2-1,0%-36-3; dan 1 mutan putatif warna orange yaitu M2-1,0%-36-1. Metode aplikasi kronik T. erecta generasi M2 menghasilkan 10 mutan putatif warna putih yaitu M2-1/10-08-2, M2-1/10-15-2, M2-1/10-23-2, M2-1/10-23-4, M2-1/10-25-4, M2-1/100-18-3, M2-1/10-25-2, M2-1/1000-02-1, M2-1/1000-15-4, dan M2-1/1000-29-4. Generasi M2 T. erecta juga menghasilkan 3 mutan putatif warna kuning yaitu M2-1/10-16-1, M2-1/10-16-2, dan M2-1/10-16-4. Marigold is one of the ornamental plant horticultural commodities that are widely planted in Indonesia. People know this plant because of its pungent aroma, known as the flower "tahi kotok" or "telek-telekan" in Javanese and "gumitir" flower in Balinese. The diversity of Tagetes sp. still relatively low, therefore, to increase the diversity of marigold plants can be done through chemical induction mutations with EMS (Ethyl Methane Sulfonate). In this study, the authors applied acute and chronic EMS chemical mutagens to Tagetes sp. In acute application method, chemical mutagen is given once, with a high concentration level so that the plant's ability to survive, and the number of mutagens produced is lower. As an alternative to increase mutant viability, the method of chronic application of EMS mutagens is to provide low levels of EMS concentration by diluting the EMS concentration at LC50 and soaking the seeds for a longer period of time.
This study aims to obtain the LC50 (Lethal Concentration 50) value, determine the sensitivity level of plants, and obtain plant diversity of Tagetes sp. with EMS acute and chronic application methods. The genetic material used in this study was derived from T. erecta genotype MG04 and T. patula genotype MG21. The acute mutation technique was performed by soaking the seeds in EMS solution for 4 hours under 10 different concentration levels (0%; 0,2%; 0,4%; 0,6%; 0,8%; 1,0%; 1,5%; 2,0%; and 3,0%). The chronic mutation technique was carried out by diluting the concentration of EMS LC50 1/10x, 1/100x, 1/1000x and immersion time of 6 hours, 24 hours and 48 hours. Observations were made on the percentage of surviving plants, quantitative and qualitative characters. The results showed that the LC50 value of T. erecta was 0,82% and LC50 of T. patula is 1,87%, which means that the sensitivity level of T. erecta to EMS treatment is quite higher than T. patula. The acute application method shows a relatively low mutation rate and limited diversity of flower types. The chronic application method produced various flower shapes and whiter flower color in T. erecta and T. patula resulted in a redder flower color than the acute application method.
The acute application method of T. erecta generation M2 produced 4 white putative mutant plants, namely M2-0,2%-20-1; M2-0,4%-39-1; M2-0,6%-20-2; M2-0,8%-20-2. There were 2 yellow putative mutants, namely M2-1,0%-36-2; M2-1,0%-36-3, and 1 orange putative mutant, namely M2-1,0%-36-1. The chronic application method of T. erecta generation M2 produced 10 white putative mutants, namely M2-1/10-08-2, M2-1/10-15-2, M2-1/10-23-2, M2-1/10 -23-4, M2-1/10-25-4, M2-1/100-18-3, M2-1/10-25-2, M2-1/1000-02-1, M2-1/1000-15-4, and M2-1/1000-29-4. The M2 generation of T. erecta also produced 3 yellow putative mutants, namely M2-1/10-16-1, M2-1/10-16-2, and M2-1/10-16-4.
