      Profil Eritrogram Sampel Darah Berantikoagulan Tripotassium Ethylene Diamine Tetraacetic Acid (K3EDTA) yang Disimpan Selama 48 Jam pada Suhu 4˚C

      Abstract
      Hasil pemeriksaan hematologi dipengaruhi oleh beberapa variabel preanalisis, termasuk antikoagulan yang digunakan, suhu penyimpanan, dan waktu antara sampel darah dikoleksi dan dianalisis. Analisis sampel darah yang tertunda karena penyimpanan yang lama dapat memengaruhi interpretasi data yang dihasilkan dari profil eritrogram. Penelitian ini bertujuan untuk mempelajari profil eritrogram pada sampel darah yang diberi antikoagulan tripotassium ethylene diamine tetraacetic acid (K3EDTA) dan disimpan selama 48 jam pada suhu 4˚C. Penelitian ini menggunakan sampel darah yang berasal dari 10 ekor sapi peranakan ongole yang sehat secara klinis. Sampel darah diambil dari vena jugularis atau coccygeal menggunakan tabung vacutainer K3EDTA, kemudian dibawa ke laboratorum dan disimpan pada suhu 4˚C. Pemeriksaan hematologi dilakukan pada 0 jam (kontrol, ± 1,5 jam setelah koleksi darah) dan pada 3, 6, 9, 12, 24, dan 48 jam setelah penyimpanan pada suhu 4˚C. Analisis profil eritrogram meliputi jumlah eritrosit, kadar hemoglobin, nilai hematokrit, dan morfologi eritrosit. Hasil pemeriksaan menunjukkan adanya peningkatan jumlah eritrosit dan nilai hematokrit (P<0,05), masing-masing pada 9 hingga 24 jam dan pada 6 hingga 48 jam setelah penyimpanan. Terdapat pula perbedaan yang signifikan (P<0,05) antara waktu penyimpanan 0 jam dengan waktu penyimpanan lainnya pada ukuran (diameter) eritrosit. Kelainan morfologi (bentuk) eritrosit berupa ekinosit terlihat pada penyimpanan 0 jam dan terus meningkat hingga 48 jam penyimpanan. Adanya hipokromasia juga ditemukan pada 6 hingga 48 jam setelah penyimpanan. Pemeriksaan hematologi untuk parameter eritrogram harus dilakukan sesegera mungkin, sebaiknya tidak lebih dari 3 jam setelah pengambilan sampel darah untuk validitas hasil.
       
      Haematological results are influenced by several pre-analytical variables, including anticoagulants used, the storage temperature, and the time between blood samples were taken and analysed. Delayed sample analysis due to prolonged storage could result in erythrogram profiles, which could complicate the interpretation of the resulting data. This experiment aimed to study the erythrogram’s profile of tripotassium ethylene diamine tetra acetic acid (K3 EDTA) bood samples stored for 48 hours at 4˚C. A total of ten healthy blood donors (Ongole crossbreed cattle) were used for this study. Blood samples were collected into K3EDTA tubes from jugular or coccygeal veins and analysed for erythrogram profiles (erythrocyte counts, hemoglobin levels, and hematocrit value, and erythrocyte morphology). Blood sample analysis for control (0 hours) was done within ± 1,5 hours after collection; they were then stored refrigerated (4˚C) and analysed from 3, 6, 9, 12, 24 and 48 hours. The results showed that there were an increased (P<0,05) on erythrocyte counts and hematocrit values at 9 to 24 hours and at 6 to 48 hours of storage, respectively. There was a significant difference between 0 hours and other time observations on the erythrocyte’s diameter (P<0,05). Morphological abnormalities of erythrocytes (echinocytes) were observed at 0 hours of storage and continued to increase up to 48 hours. Hypochromasia also found at 6 hours to 48 hours of storage. Therefore, analysis of blood samples for erythrogram parameters should be carried out as soon as possible, preferably within 3 hours after their collection to ensure clinically reliable results.
