Pestisida merupakan zat atau senyawa kimia yang digunakan untuk melindungi tanaman, mencegah, memusnahkan, menolak atau memusuhi hama dalam bentuk hewan, tanaman, dan mikroorganisme pengganggu sehingga penggunaanya mampu meningkatkan hasil pertanian. Penggunaan pestisida merupakan bagian yang tidak dapat terpisahkan dari budidaya pertanian, di Indonesia sendiri sebanyak 95,29% petani menggunakan pestisida kimia sebagai sarana dalam mengendalikan organisme penggangu tanaman. Formula pestisida berdsarkan bahan aktifnya yang terdaftar pada tahun 2016 sebanyak 3.207 formula, dan pada tahun 2020 mengalami peningkatan menjadi 3.525. Klorpirifos merupakan insektisida organofosfat yang paling banyak digunakan untuk mengendalikan serangga pada tanaman sayuran dan buah. Insektisida tersebut cukup beracun karena memiliki sifat neurotoksik dan imunotoksik yang berbahaya untuk hewan dan manusia. Selain itu, klorpirifos memiliki tingkat kelarutan yang sangat rendah yaitu 1,4 mg kg-1 sehingga residunya bertahan lebih lama di dalam tanah, dapat terserap oleh tanaman dan mencemari lingkungan. Residu insektisida klorpirifos yang semakin meningkat di dalam tanah dapat mempengaruhi kesuburan tanah dan mengganggu keseimbangan lingkungan. Bioremediasi merupakan teknologi yang digunakan untuk mengatasi masalah pencemaran. Teknologi tersebut dilaporkan lebih murah, efektif, dan ramah lingkungan. Bakteri merupakan salah satu mikrob yang banyak digunakan untuk bioremediasi karena siklus hidupnya pendek dan pertumbuhannya dapat dioptimasi. Tujuan dari penelitian ini adalah untuk mengisolasi dan mengkarakterisasi bakteri yang mampu mendegradasi pestisida berbahan aktif klorpirifos serta mempelajari mekanisme bakteri dalam mendegradasi bahan aktif tersebut
Bakteri diisolasi dari sampel rhizosfir dan endofit tanaman bawang merah asal Kabupaten Brebes dan Kabupaten Sumbawa. Isolat bakteri yang diperoleh diskrinning resistensinya terhadap pestisida klorpirifos, profenofos, glifosat, dan paraquat. Isolat bakteri yang mempunyai resistensi tinggi terhadap pestisida uji dikarakterisasi dan diidentifikasi secara molekuler sampai tingkat spesies. Karakterisasi dilakukan terhadap bakteri yang mempunyai resistensi tinggi meliputi uji hipersensitivitas pada daun tembakau, uji hemolisis pada media agar darah, uji kompatibilitas antarbakteri, pembuatan kurva baku dan kurva tumbuh, serta kemampuan fungsional bakteri dalam menghasilkan hormone tumbuh IAA. Mekanisme bakteri mendegradasi klorpirifos diamati melalui uji pemanfaatan klorpirifos sebagai sumber karbon, fosfat, dan keduanya. Pengamatan optimasi biodegradasi bakteri secara in vitro meliputi pengaruh pemberian nutrisi dan nilai pH.
Sebanyak 27 isolat bakteri berhasil diisolasi dari rhizosfir tanah dan tanaman bawang merah yang ditanampada tanah yang mengandung residu klorpirifos di atas ambang batas yaitu 0,46-0,67 mg/l. Dua isolat bakteri yaitu WS2.1 dan WU2.1 mempunyai resistensi tertinggi terhadap pestisida yaitu konsentrasi klorpirifos 30.000-38.000 ppm, gliposat 20.000 ppm, profenofos 20.000 ppm dan paraquat
8.000-10.000 ppm. Kedua isolat bakteri tidak bersifat patogen baik padatumbuhan maupun hewan dan tidak bersifat antagonis sehingga dapat dikonsorsium. Isolat bakteriWS2.1 mampu menghasilkan hormone tumbuh IAA sebesar 0,006 ppm dan biakan konsorsium sebesar 0,1 ppm. Bakteri mendegradasi klorpirifos dengan cara memanfaatkannya terutama sebagai sumber P dan sebagai sumber C. IsolatWS2.1 diidentifikasi sebagai Klebsiella pneumoniae WS2.1 (OM883979) dan isolat WU2.1 sebagai Bacillus oceanisediminis WU2.1 (OM956173). B. oceanisediminis WU2.1 mampu mendegradasi klorpirifos sebesar 98% selama 48 jam dalammedia yang tidak diberikan glukosa. Pada media dengan pH 7, K. pneumoniaeWS2.1 dan
B. oceanisediminis WU2.1 mampu mendegradasi klorpirifos sebesar 99,78 % dan 99,70% selama 48 jam. Pesticides are substances or chemical compounds used to protect plants, prevent, destroy, reject or antagonize pests in the form of animals, plants, and nuisance microorganisms so that their use can increase agricultural yields. The use of pesticides is an inseparable part of agricultural cultivation, in Indonesia alone as many as 95.29% of farmers use chemical pesticides as a means of controlling plant- disturbing organisms. In addition, chlorpyrifos has a very low solubility level of 1.4 mg kg-1 so that the residue lasts longer in the soil, can be absorbed by plants and pollute the environment. The increasing residue of chlorpyrifos insecticide in the soil can affect soil fertility and disturb the environmental balance. Bioremediation is a technology used to solve pollution problems. The technology is reported to be cheaper, more effective, and environmentally friendly. Bacteria are one of the microbes that are widely used for bioremediation because their life cycle is short and their growth can be optimized. The purpose of this study was to isolate and characterize bacteria capable of degrading pesticides with the active ingredient chlorpyrifos and to study the mechanism of bacteria in degrading the active ingredient.
Bacteria were isolated from rhizosphere and endophytic samples of shallot plants from Brebes Regency and Sumbawa Regency. The bacterial isolates obtained were screened for resistance to the pesticides chlorpyrifos, profenofos, glyphosate, and paraquat. Bacterial isolates that had high resistance to the tested pesticides were characterized and identified molecularly to the species level. Characterization was carried out on bacteria that had high resistance including hypersensitivity test on tobacco leaves, hemolysis test on blood agar media, compatibility test between bacteria, making standard curves and growth curves, as well as the functional ability of bacteria to produce growth hormone IAA. The mechanism of bacteria degrading chlorpyrifos was observed by using chlorpyrifos as a source of carbon, phosphate, and both. Observations on the optimization of bacterial biodegradation in vitro include the effect of nutrition and pH value.

Two bacterial isolates namely WS2.1 and WU2.1 had the highest resistance to pesticides, namely concentrations of chlorpyrifos 30,000-38,000 ppm, glyphosate 20,000 ppm, profenofos 20,000 ppm and paraquat 8,000-10,000 ppm. The two bacterial isolates were not pathogenic in either plants or animals and were not antagonistic so they could be consorted. The bacterial isolate WS2.1 was able to produce growth hormone IAA of 0.006 ppm and the consortium culture was able to produce growth hormone of 0.1 ppm. Bacteria degrade chlorpyrifos by utilizing it mainly as a source of P and as a source of C. The isolate WS2.1 was identified as
Klebsiella pneumoniae WS2.1 (OM883979) and isolate WU2.1 as Bacillus oceanisediminis WU2.1 (OM956173). B. oceanisediminis WU2.1 was able to degrade chlorpyrifos by 98% for 48 hours in media that was not given glucose. In media with pH 7, K. pneumoniae WS2.1 and B. oceanisediminis WU2.1 were able to degrade chlorpyrifos 99.78% and 99.70% for 48 hours, respectively.
