      Depolimerisasi Kitosan Karapas Udang dengan Enzim Papain dan Sinar Ultraviolet

      Abstract
      Kitosan yang beredar di pasar Indonesia memiliki karakteristik berat molekul besar dan hanya larut dalam pH asam, sehingga pemanfaatannya terbatas. Depolimerisasi kitosan menjadi oligokitosan dihasilkan produk dengan karakteristik berat molekul rendah dan mudah larut dalam pH netral, sehingga pemanfatannya luas. Tujuan penelitian ini adalah menentukan metode depolimerisasi kitosan karapas udang untuk menghasilkan oligokitosan dengan menggunakan enzim papain (pH 5, 40 °C, 16 jam) dan sinar ultraviolet (75 menit), serta mengkarakterisasi kitosan dan oligokitosan karapas udang yang dihasilkan dari metode depolimerisasi yang berbeda. Aktivitas pada penelitian ini meliputi depolimerisasi kitosan dengan menggunakan enzim papain dan sinar ultraviolet, serta karakterisasi oligokitosan. Oligokitosan dari kitosan pharmaceutical grade berhasil diperoleh dengan menggunakan ketiga metode depolimerisasi tersebut. Metode depolimerisasi kombinasi (sinar ultraviolet dan enzim papain) menghasilkan oligokitosan terbaik dengan rendemen 66,00±1,80%, derajat putih 99,33±0,76%, kelarutan 100%, viskositas 92,13±0,61 cP, berat molekul 4,94±0,02 kDa, dan derajat deasetilasi 90,40%.
       
      Chitosan circulating in the Indonesian market has the characteristics of a large molecular weight and is only soluble at an acidic pH, so its utilization is limited. The depolymerization of chitosan into oligochitosan produces products with characteristics of low molecular weight and easily soluble at neutral pH so that the utilization is wide. The purpose of this study was to determine the depolymerization method of shrimp carapace chitosan to produce oligochitosan using papain enzymes (pH 5, 40 °C, 16 hours) and ultraviolet light (75 minutes), and to characterize the chitosan and shrimp carapace oligochitosan produced by different depolymerization methods. Activities in this study include depolymerization of chitosan using papain enzymes and ultraviolet light, and characterization of oligochitosan. Oligochitosan from pharmaceutical grade chitosan was successfully obtained using the three depolymerization methods. The combination depolymerization method (ultraviolet light and papain enzyme) produced the best oligochitosan with a yield of 66,00±1,80%, whiteness 99.33±0,76%, solubility 100%, viscosity 92,13±0,61 cP, molecular weight 4,94 ±0,02 kDa, and the degree of deacetylation was 90,40%.
