Pencemaran limbah merkuri di dalam tanah terjadi karena beberapa akibat, antara lain pembuangan limbah industri yang tidak melalui pengolahan, penggunaan merkuri dalam pertanian, serta limbah hasil penambangan atau pengolahan emas. Toksisitas atau daya racun logam berat tergantung dari jenis, efek, kadar, antagonis atau sinergi, dan sifat kimianya. Semakin besar kadar logam berat maka daya toksisitasnya semakin besar. Jumlah Hg di dalam tanah sebesar 0,3 mg/L untuk nilai baku karakteristik beracun, sedangkan untuk total konsentrasi sebesar 300 mg/kg merupakan penetapan pengelolaan tanah terkontaminasi limbah bahan berbahaya dan beracun. Logam berat Hg sangat sulit terdegradasi sehingga kadar Hg dalam tanah di areal pertambangan yang sudah berjalan beberapa tahun menjadi tinggi. Hal ini menjadi masalah dan fokus utama apabila logam
Hg tersebut masuk dan mengendap di dalam tanah dan air yang kemudian dapat masuk ke rantai makanan makhluk hidup.
Penanggulangan masalah pencemaran akibat akumulasi logam berat Hg perlu dilakukan suatu usaha pembersihan (clean up) atau yang disebut remediasi dari bekas tempat penimbunan limbah yang mengandung bahan berbahaya dan beracun. Remediasi yang dilakukan secara biologi atau disebut bioremediasi merupakan pengembangan dari bidang bioteknologi lingkungan dengan memanfaatkan proses biologi dalam mengendalikan pencemaran. Pada penelitan ini bertujuan untuk mendapatkan kultur fungi
yang berpotensi sebagai penyerap logam berat Hg. Hasil penelitian ini diharapkan agar teknologi bioremediasi dengan menggunakan fungi dapat diterapkan sebagai alternatif metode pemulihan tanah terkontaminasi logam Hg yang ramah lingkungan.
Pada penelitian ini sampel tanah tercemar logam berat Hg diambil dari limbah tambang emas milik masyarakat di Desa Kertajaya, Kecamatan Simpenan, Kabupaten Sukabumi (7°06'11.3"S 106°35'48.8"E). Tahapan penelitian meliputi isolasi fungi, purifikasi dan morfologi isolat fungi, uji hipersensitivitas dan patogenisitas, skrining isolat fungi, skrining
kualitatif isolat fungi untuk aktivitas ligninolitik (lakase), identifikasi molekuler, uji kompatibilitas antarfungi, dan efektivitas fungi resisten Hg secara in vitro.
Hasil penelitian menunjukkan bahwa total fungi yang didapatkan dari sampel tailing dan rhizosfer 23 isolat. 3 isolat fungi potensial berkode M.2, M.10, dan M.13 mampu tumbuh dengan konsentrasi merkuri 350 ppm, sedangkan 1 isolat berkode R4.28 mencapai konsentrasi 1000 ppm. Selain itu, isolat M.2 terpilih sebagai galur terbaik dibandingkan dengan isolat M.10, M.13, dan R4.28 untuk uji kualitatif produksi lakase pada enzim
ligninolitik. Hasil analisis molekuler gen 18S rRNA, isolat M.2 memiliki kemiripan dengan Trichoderma yunnanense dengan nilai similarity dari hasil BLAST sebesar 98,49%, sedangkan isolat R4.28 memiliki kemiripan dengan Penicillium soli sebesar 99,32%. Fungi Penicillium soli R4.28 mampu menurunkan konsentrasi Hg tertinggi yang mencapai 93,7%, sedangkan Trichoderma yunnanense M.2 mampu menurunkan konsentrasi Hg sebesar 92,7% dalam waktu 7 hari setelah inkubasi dengan penurunan pH mencapai 4,6 - 4,7 dari pH awal yaitu 6,8. Penurunan pH oleh isolat fungi dapat disebabkan karena naiknya kadar asam pada sampel sebagai hasil dari proses pengikatan gugus-gugus fungsional selama proses inkubasi. Mercury waste pollution in the soil occurs due to several consequences, including the disposal of industrial waste that does not go through processing, the side of the use of mercury compounds in agriculture, as well as waste resulting from mining or processing of gold. The toxicity or toxic power of heavy metals depends on their type, effect, content, antagonist or synergy, and chemical properties. The amount of Hg in the soil is 0.3 mg/L for the raw value of toxic characteristics, while for the total concentration of 300 mg/kg is the determination of soil management contaminated with hazardous and toxic waste materials. Heavy metals of Hg are so difficult to degrade that Hg levels in the soil in mining areas that have been running for several years are high. This becomes the main problem and focuses when the Hg metal enters and settles in the soil and water which can then enter the food chain of living things.
Tackling pollution problems due to the accumulation of heavy metals Hg needs to be done a cleanup effort or so-called remediation of the former landfill containing hazardous and toxic materials. Remediation conducted biologically or called bioremediation is a development of the field of environmental biotechnology by utilizing biological processes in controlling pollution. This research, it's aimed at obtaining a functional culture that is
potentially a heavy metal absorber Hg. So it is hoped that bioremediation technology using functions can be applied as an alternative method of soil recovery contaminated environmentally friendly Hg metal.
In this research, soil samples contaminated with heavy metal Hg were taken from gold mining owned by the people in Kertajaya Village, Simpenan District, Sukabumi Regency. The stages of the research include isolation of fungi, purification and morphology of fungal isolates, hypersensitivity and pathogenicity tests, screening of fungal isolates, qualitative
screening of fungal isolates for ligninolytic activity (laccase), molecular identification, inter-fungal compatibility test, and the effectiveness of Hg resistance fungi in vitro.
The results showed that the total fungi obtained from tailings and rhizosphere samples were 23 isolates. 3 potential fungal isolates coded M.2, M.10, and M.13 were able to grow at a mercury concentration of 350 ppm, while 1 isolate coded R4.28 reached a concentration of 1000 ppm. In addition, isolate M.2 was selected as the best strain compared to isolates M.10, M.13, and R4.28 for a qualitative test of laccase production in ligninolytic enzymes. The results of the molecular analysis of the 18S rRNA gene, isolate M.2 had similarities with Trichoderma yunnanense with a similarity value from the BLAST results of 98.49%,
while isolate R4.28 had similarities with Penicillium soli of 99.32%. Fungi Penicillium soli R4.28 was able to reduce the highest Hg concentration reaching 93.7%, while Trichoderma yunnanense M.2 was able to reduce Hg concentration by 92.7% within 7 days after incubation with a decrease in pH reaching 4.6 - 4.7 from the initial pH of 6.8. The decrease in pH by fungal isolates could be caused by the increase in acid levels in the sample as a
result of the binding process of functional groups during the incubation process.
