Sapi pasundan salah satu plasma nutfah sapi lokal Indonesia yang perlu dikembangkan dan dilestarikan. Sapi pasundan merupakan hasil persilangan antara Bos javanicus (sapi bali) dan Bos indicus (sapi ongole dan sapi madura). Perbaikan mutu genetik sapi pasundan diupayakan oleh pemerintah melalui program inseminasi buatan (IB) menggunakan semen beku. Pemilihan pejantan unggul melalui breeding soundness examination (BSE) mensyaratkan kualitas semen baik. Kualitas semen ditentukan oleh usia ketika hewan tersebut telah mencapai pubertas.
Prediksi fertilitas pejantan belum cukup akurat berdasarkan penilaian kualitas semen menggunakan parameter konvensional. Pengujian secara molekuler menjadi salah satu upaya dalam mendeteksi kualitas sperma. Teknik seleksi secara genomik terhadap kualitas semen yang cepat, tepat, dan akurat belum dikembangkan. Gen yang diekspresikan terkait dengan kualitas semen belum banyak dilaporkan dan perlu dilakukan konfirmasi, khususnya sapi pasundan belum pernah dilaporkan. Penelitian ini dilakukan untuk mengembangkan metode deteksi molekular dalam penilaian kualitas semen sapi pejantan.
Penelitian terdiri atas 4 bagian. Penelitian bagian pertama bertujuan untuk melakukan kajian terhadap morfometrik tubuh serta korelasinya dengan konsentrasi testosteron pada kelompok sapi jantan prepubertas berumur <6 bulan (I), peri-pubertas berumur 6-12 bulan (II), post-pubertas (III) dan pejantan (IV) sapi pasundan berumur >12 bulan. Hasil penelitian menunjukkan lebar dada, tinggi panggul, panjang kepala, dan lebar kepala berbeda (P<0,05) antara kelompok I dengan II. Parameter morfomotrik seluruhnya berbeda nyata (P<0,05) pada kelompok III dan IV. Persentase peningkatan rerata morfometrik tubuh sebesar 59,05% (fase prepubertas ke peri-pubertas) dan 71,38% (fase peri-pubertas ke post-pubertas). Indeks morfometrik height slope, length index (2), depth index, dan foreleg length berbeda (P<0,05) antara III dengan IV. Rerata konsentrasi hormon testosteron berbeda (P<0,05) antara kelompok I (3,25±0,76 ng ml-1), II (9,13±0,85 ng ml-1) dan III (12,16±0,40 ng ml-1). Umur berkorelasi (P<0,05) dengan lingkar skrotum pada fase prepubertas (R² = 0,9973) dan post-pubertas (R² = 0,4150), tetapi tidak dapat dijadikan sebagai penentu kadar testosteron.
Penelitian bagian kedua bertujuan untuk mengevaluasi karakteristik tubuh, libido, dan konsentrasi testosteron serta korelasinya antar parameter pada sapi jantan pasundan. Sapi pasundan dikelompokkan berdasarkan motilitas sperma semen segar ke dalam kelompok A (70-79%) dan B (80-89%). Hasil penelitian menunjukkan fenotipik dan umur tidak berbeda (P>0,05) antara kelompok A dan B. Perbedaan morfometrik (P<0,05) dijumpai pada lebar kepala (A: 23,04±0,24 cm dan B: 21,60±0,60 cm). Rerata konsentrasi testosteron sebesar 13,38±0,21 ng ml-1. Waktu consummation kelompok A (108,20±20,16 sa) lebih cepat (P<0,05) dibandingkan dengan kelompok B (184,60±26,66 sa). Panjang badan berkorelasi dengan waktu courtship sebesar (R2=0,9596) dan consummation (R2=0,9487) pada kelompok A. Disimpulkan bahwa konsentrasi testosteron, umur, dan lingkar skrotum tidak dapat digunakan untuk menduga motilitas sperma.
Penelitian ketiga bertujuan untuk mengevaluasi karakteristik semen dan potensi produksi semen beku pejantan sapi pasundan. Hasil penelitian menunjukkan pH semen segar kelompok B lebih rendah (P<0,05) dibandingkan dengan kelompok A. Konsentrasi sperma per ejakulat pada kedua kelompok tidak berbeda (P>0,05) dengan kisaran 4312,36x106 - 6303,52 x106 sperma. Abnormalitas, viabilitas dan keutuhan DNA sperma semen segar tidak berbeda (P>0,05) antara kelompok A (9,41±1,21%; 84,41±0,99%; 91,19±0,79%) dan kelompok B (10,22±0,66%; 86,35±2,16%; 92,58±0,35%). Keutuhan DNA sperma semen beku kelompok B (89,81±1,18%) lebih tinggi (P<0,05) dibandingkan dengan kelompok A (86,83±0,60%). Motilitas sperma semen beku kelompok A (<40%) lebih rendah dibandingkan dengan kelompok B. Parameter kinematik motilitas sperma yaitu curvilinear velocity (VCL), straight-line velocity (VSL), average pathway velocity (VAP), amplitude of lateral displacement (ALH), dan beat cross frequency (BCF) tidak berbeda (P<0,05) antara kelompok A dan B. Produksi semen beku pejantan pasundan antara 144,18 sampai 191,29 straw/ejakulat. Penelitian disimpulkan bahwa kelompok B memenuhi syarat sebagai sumber semen untuk dibekukan dan digunakan dalam IB.
Penelitian bagian keempat bertujuan mengisolasi RNA dari sperma serta mempelajari profil ekspresi gen protamin 1 (PRM1), protamin 2 (PRM2) dan deleted in azoospermia-like (DAZL). Hasil transkrip PRM1, PRM2, DAZL, dan ß-Actine (ACTB) diperlihatkan dalam sperma menggunakan primer PCR yang sesuai. Ekspresi gen PRM1, PRM2, dan DAZL dievaluasi menggunakan qRT-PCR dan ACTB digunakan sebagai kontrol. Motilitas progresif sperma semen segar 80-89% memperlihatkan tingkat ekspresi mRNA PRM1 dan PRM2 secara signifikan lebih tinggi (P<0,05) dibandingkan dengan motilitas progresif sperma 70-79%. Hal ini menunjukkan adanya peran gen terhadap parameter motilitas progresif sperma.
Hasil kajian penanda genetik mampu menjelaskan gen yang berperan dalam kualitas sperma. Gen yang dikaji merupakan penanda genetik spesifik dan metode akurat untuk seleksi pejantan. Ekspresi gen PRM1 dan PRM2 dengan tingkat ekspresi relative >1,0 dapat digunakan sebagai parameter biomulekular dalam seleksi pejantan unggul sapi pasundan. Pasundan cattle are one of the Indonesian domestic resources of genetic bull that need to be developed and conserved. Pasundan cattle initially came from crossbreeding between Bos javanicus (Bali cattle) and Bos indicus (Ongole and Madura cattle). The government is trying to improve the genetic quality of Pasundan cattle through an artificial insemination (AI) program using frozen semen. Bull’s selection through breeding soundness examination (BSE) requires good quality of semen. Semen quality is determined by males when they reach puberty.
Prediction of bulls fertility is not accurate enough based on quality assessment of semen using conventional parameters. Molecular testing is an effort to detect sperm quality. The rapidly precision and accuracy of genomic selection techniques in semen quality is limited. Genes expression related to semen quality have not been widely reported and need to be confirmed, especially in Pasundan cattle. This study aimed to develop a molecular detection method for assessing semen quality in Pasundan bulls.
This study consists of 4 experiments. The first experiment aims to study the morfometric characteristics of body measurements and their relationship with testosterone concentration in pre at <6 months of age (I), peri at 6-12 months of age (II), post-pubertal at >12 months of age (III) and Pasundan mature bulls at >12 months of age (IV). The results showed that hearth girth, rumpt height, and head were significantly different (P<0.05) between group I and II. All morphometric parameters were significantly different (P<0.05) between group III and IV. Body morphometric increases from prepubertal to peri-puberty phase was 59.05% and from peri-puberty to postpuberty phase was 71.38%. Morphometric index height slope, length index (2), depth index, and foreleg length was significantly different (P<0.05) between group III and IV. The value of testosterone concentration was significantly different (P<0.05) between groups I (3.25±0.76 ng ml-1), II (9.13±0.85 ng ml-1) and III (12.16±0.40 ng ml-1). Age had correlation (P<0.05) with scrotal circumference in prepubertal (R² = 0.9973) and post-pubertal (R² = 0.4150) phase, however, cannot use as testosterone levels determine.
The second experiment aimed to evaluate phenotypic, morphometric, libido, and concentration of testosterone and their relationship between parameters in Pasundan bulls. Pasundan bulls were divided into two groups based on their sperm motilities into group A (70-79%) and group B (80-89%). The results showed that phenotype and age did not different (P>0.05) between groups A and B. The morphometric variations were significantly different (P<0.05) on face width (A: 23.04±0.24 cm and B: 21.60±0.60 cm). The value of testosterone concentration was 13.38±0.21 ng ml-1. Consummation time in group A (108.20±20.16 sa) was faster (P<0.05) than group B (184.60±26.66 sa). Body length was correlated with courtship (R2=0.9596) and consummation time (R2=0.9487) in group A. In conclusion, testosterone concentration, age, and scrotal circumference can not used for predicting of sperm motility.
The third experiment aimed to evaluate the sperm characteristics and frozen semen production potential in Pasundan bulls. The results showed that the fresh semen pH in group B was lower (p<0.05) than group A. Sperm concentration per ejaculate showed no difference (p>0.05) with a range of 4312.36x106 to 6303.52x106. The viability and DNA integrity of fresh semen were not different (p>0.05) between group A (84.41±0.99%; 91.19±0.79%) and group B (86.35±2.16%; 92.58±0.35%). DNA integrity of frozen semen of group B (89.81±1.18%) was higher (p<0.05) than group A (86.83±0.60%). The frozen semen sperm motility in group A (<40%) was lower than group B. However, the sperm motility kinematic parameters of curvilinear velocity (VCL), straight-line velocity (VSL), average pathway velocity (VAP), amplitude of lateral displacement (ALH), and beat cross frequency (BCF) were not different (p<0.05) between two groups. The frozen semen production of Pasundan bulls were between 144.18 to 191.29 straws/ejaculate. In conclusion, group B are eligible to be used as a semen source for freezing to be use in AI program.
The fourth experiment aimed to isolate RNA from mature sperm and to monitor the expression profile of protamine 1 (PRM1), protamine 2 (PRM2) and deleted in azoospermia-like (DAZL) gene. The results showed that the levels of PRM1 and PRM2 mRNA expression in progressive motility of fresh semen 80-89% was higher (P<0.05) than progressive motility of fresh semen 70-79%. This indicates the role of genes in semen quality parameters.
The results of the genetic markers study can explain the genes that play a role in sperm quality. The analysis of genes was a specific genetic marker and accurate method for the selection of bulls. The expression of PRM1 and PRM2 genes with relative expression levels > 1.0 could use as biomolecular parameters for selecting pasundan bulls.
