      Rasio Neutrofil/Limfosist Anak Sapi yang Diinfeksi dengan Bakteri Escheria coli K99 dan Diberi Mikrokapsul IgG Anti Escheria coli

      Abstract
      Stres merupakan respon biologis yang dapat menimbulkan ancaman dan mengganggu homeostasis pada hewan. Stres dapat diketahui melalui pengamatan terhadap indikator stres diantaranya rasio neutrofil terhadap limfosit (rasio N/L). Penelitian ini bertujuan untuk mengetahui profil rasio N/L sebagai indikator stres pada anak sapi Friesian Holstein yang diinfeksi bakteri Eschericia coli K-99 dan diberi mikrokapsul anti E. coli. Dua puluh ekor anak sapi neonatus sehat secara klinis, dibagi menjadi lima kelompok (masing-masing kelompok empat ekor), terdiri dari kelompok KN (kontrol negatif; tidak diinfeksi, tidak diberi perlakuan); kelompok KP (kontrol positif; diinfeksi; tanpa perlakuan); kelompok perlakuan AB (diinfeksi dan diberi antibiotik); kelompok perlakuan KOL (diinfeksi dan diberi suspensi IgG anti E.coli); dan kelompok perlakuan MK (diinfeksi dan diberi mikrokapsul IgG anti E.coli). Semua anak sapi kecuali kelompok kontrol negatif diinfeksi E. coli sebanyak 5 x 1010 colony forming unit/ekor secara oral. Antibiotik diberikan secara intramuskular dengan dosis 6mL/50 kgBB/hari selama tiga hari. Sediaan suspensi IgG anti E. coli dan mikrokapsul IgG anti E. coli masing-masing diberikan secara oral sebanyak 1.76 g IgG anti E. coli/ekor, pada 1 jam sebelum diinfeksi, 4 dan 9 jam setelah infeksi dan dilanjutkan dua kali sehari masing-masing pada dua hari berikutnya. Sampel darah diambil dari vena jugularis externa pada saat sebelum infeksi dan pada 12, 24, 48 dan 72 jam setelah infeksi untuk dianalisis terhadap jumlah neutrofil dan limfosit serta rasio neutrofil terhadap limfosit (rasio N/L). Hasil pengamatan menunjukkan bahwa rasio N/L anak sapi yang diinfeksi dengan E. coli K-99 dan diberi mikrokapsul IgG anti E. coli tidak berbeda nyata, baik antar kelompok maupun antar waktu pengamatan (P>0.05). Infeksi dengan E. coli K-99 dan pemberian mikrokapsul IgG anti E. coli pada anak sapi tidak memengaruhi indeks stres (rasio N/L).
       
      Stress is a biological response which may affect the homeostasis of animal body. Stress can be recognised through observation of certain indicators including neutrophil to lymphocyte ratio. The objective of this experiment was to study the neutrophil to lymphocyte ratio (N/L ratio) profiles of calves infected by Escherichia coli and administered by anti E. coli IgG microcapsules. Twenty calves, clinically healthy, divided into five groups, consist of five calves each group, i.e. negative control/KN group (not infected and no treatments); positive control/KP group (infected and no treatments); AB group (infected; antibiotic treatments); KOL group (infected and suspension of anti E. coli IgG treatments); and MK group (infected and anti E. coli IgG microcapsules treatments). All the calves except the negative control group induced E. coli as much as 5 times 1010 colony forming unit orally. The E. coli K-99 was administered orally with the dosage of 5 × 1010 cfu/ml per calf. Antibiotic was given intramuscularly at a dose of 6 mg/50 kg BW/day for three days. The suspension and microcapsules of anti E. coli K-99 IgG were administered orally with the dosage of 1.76 gram anti E. coli K-99 IgG/calf which is given at 1 hour before, 4 and 9 hours after infection on the first day and twice a day for the following two consecutive days. The blood samples were collected from jugular vein at 0, 12, 24, 48, and 72 after E. coli K99 infection, for neutrophil and lymphocyte, and neutrophil to lymphocyte ratio (N/L ratio) analysis. Results of this experiment showed that there were no significantly different between treatments and also between the time of examination (P>0.05). The infection of calves with Escherichia coli and administered by anti E. coli IgG microcapsules didn’t affect the stress index (N/L ratio).
