Analisis asam nukleat berpotensi sangat baik untuk diaplikasikan di bidang medis, forensik, arkeologi, dan pangan. Thiazole orange (TO) dapat digunakan sebagai pewarna dan pemindai fluoresens dalam analisis asam nukleat. Penelitian ini bertujuan mengkaji senyawa turunan thiazole orange (TO-n) serta mengulas pengaruh modifikasi struktur terhadap parameter fotofisika dan potensi aplikasinya. Modifikasi senyawa TO-n dilakukan dengan penyisipan berbagai jenis substituen pada bagian benzotiazola, kuinolina, dan metina. Faktor elektronik dan ukuran molekul substituen akan mempengaruhi parameter fotofisika senyawa TO-n. Senyawa TO-11 mengalami peningkatan intensitas fluoresensi paling tinggi, yaitu sebesar >2030 kali. Keberadaan gugus stiril meningkatkan spesifisitas deteksi terhadap RNA dan G-quadruplex DNA, sedangkan gugus aromatik dan gugus beratom nitrogen berpengaruh terhadap spesifisitas deteksi DNA untai ganda (dsDNA). Gugus stiril, benzil, dan klorin dapat meningkatkan fotostabilitas senyawa TO-n terhadap oksigen singlet (1O2). Nucleic acid analysis has excellent potential for applications in the medical, forensic, archaeological, and food fields. In nucleic acid analysis, thiazole orange can be used as a dye and a fluorescent probe. This research aims to examine the thiazole orange (TO-n) derivative to review how structural changes affect photophysical parameters and potential uses in nucleic acid analysis. Different kinds of the substituent are inserted on the benzothiazole, quinoline, and methine moiety to modify the TO-n structure. The TO-11 compound exhibited a significant increase in fluorescence intensity, which was >2030 times. Electronic factors and molecular size of substituent will affect the photophysical parameters of thiazole orange derivatives. The presence of the styryl group increased the specificity of detection for RNA and G-quadruplex DNA, while the halogen group affected the specificity of dsDNA detection. Styryl, benzyl, and chlorine groups can also increase the photostability of TO-n compounds against singlet oxygen (1O2).
