      Karakterisasi Streptomyces isolat OM1CHD dan OM2RH(e) dan Aktivitas Enzim Mananasenya

      Abstract
      Streptomyces adalah golongan bakteri dari kelas Aktinomisetes yang dapat menghasilkan bentuk dan pigmen yang beragam. Streptomyces memiliki potensi dalam menghasilkan enzim mananase yang dapat menghidrolisis senyawa polisakarida manan menjadi manooligosakarida. Oleh karena itu, penelitian ini bertujuan mengkarakterisasi morfologi Streptomyces isolat OM1CHD dan OM2RH(e) serta aktivitasnya dalam memproduksi enzim mananase. Karakterisasi dilakukan dengan menumbuhkan kedua isolat pada 4 media yang berbeda yaitu ISP 2, ISP 3, ISP 4, dan HVA kemudian diamati pertumbuhannya secara makroskopis dan mikroskopis. Uji kualitatif dilakukan dengan mengamati zona bening pada media padat dan uji kuantitatif dilakukan dengan mengamati aktivitas harian enzim mananase menggunakan metode DNS. Pertumbuhan pada 4 media yang berbeda menunjukkan pertumbuhan yang berbeda pula di antara kedua isolat uji. Pengamatan mikroskopis menunjukkan kedua isolat memiliki rantai spora dengan tipe Rectiflexibiles dan menghasilkan pigmen kuning pada media ISP 2 dan brownbeige pada media HVA. Indeks mananolitik terbesar dimiliki oleh Streptomyces OM2RH(e) dengan nilai indeks mananolitik pada media BIS dan LBG adalah sebesar 0,34 dan 0,45. Aktivitas enzim mananase isolat OM2RH(e) pada substrat BIS 0.5% tertinggi terjadi pada hari ke 9 dengan nilai 0,033 U/mL.
       
      Streptomyces is a group of bacteria from the Actinomycetes class that can produce various shapes and pigments. Streptomyces has the potential to produce mannanase enzymes that can hydrolyze mannan polysaccharides into manooligosaccharides. Therefore, this study aimed to characterize Streptomyces isolates OM1CHD and OM2RH(e) and their activity in producing mannanase enzymes. Characterization was done by growing the two isolates on 4 different media, namely ISP 2, ISP 3, ISP 4, and HVA and then observed their growth macroscopically and microscopically. The qualitative test was carried out by observing the clear zone on solid media and the quantitative test was carried out by observing the daily activity of the mannanase enzyme using the DNS method. Characterization on 4 different media showed different growth between the two test isolates. Microscopic observation showed that both isolates had spore chains with the type of Rectiflexibiles and produced pigments on ISP 2 and HVA media. The largest mannanolytic index was Streptomyces OM2RH(e) with the mannanolytic index values in BIS and LBG media were 0.34 and 0.45, respectively. Mannanase enzyme activity of isolate OM2RH(e) on 0.5% BIS substrate, the highest occurred on day 9 with a value of 0.033 U/mL.
