      Abstract
      Deteksi kebuntingan dini sangat diperlukan untuk meningkatkan produksi yang efisien dan hemat biaya pada peternakan domba di Indonesia. Deteksi kebuntingan dini menggunakan barium klorida (BaCl2) memerlukan sampel dan reagen dalam volume besar dan peralatan khusus. Penelitian ini bertujuan untuk mengembangkan alat deteksi kebuntingan dini pada domba yang praktis dan ekonomis dengan melakukan penyederhanaan prinsip uji BaCl2. Perangkat mikrofluida yang dikembangkan menggunakan kain katun sebagai bahan penyerap, filter, dan tempat terjadinya reaksi. Kain katun dengan pencucian satu kali menghantarkan cairan lebih optimal. Kain katun mampu menyerap cairan sebanyak 53µL selama 10,6 detik. Kristal BaCl2 terlihat jelas pada kain katun yang mengandung urin setelah dikeringkan selama nol sampai 15 menit. Kristal BaCl2 hasil reaksi pada kain katun dapat dilihat secara langsung tanpa bantuan alat pada konsentrasi 25-50% pada menit ke sepuluh. Nilai LoD dan LoQ perangkat mikrofluida secara berurutan adalah 0,010 g/mL dan 0,031 g/mL.
       
      Early pregnancy detections are strongly needed to increase production efficiency and cost-effective production on sheep farms in Indonesia. Early pregnancy detection using barium chloride (BaCl2) requires a large volume of samples and reagent and specialized equipment. This study was aimed to develop a practical diagnostic device to detect early pregnancy in sheep by simplifying the principles of the BaCl2 test. The microfluidic device was developed using a cotton cloth as an absorbent, filter, and reaction site. The cotton cloth with single washing had proven to be more optimal in delivering fluids. The results showed that evaporation of urine affected the formation of the BaCl2 crystals. BaCl2 crystals clearly inspected at the cotton cloth that has been contained by urine after zero up to 15 minutes evaporation. The result of the BaCl2 crystallization at the cotton cloth clearly inspected by eyes without supporting tool at the concentration of 25% up to 50%. The Limit of Detection and Limit of Quantification of the microfluidic device were 0.010 g/mL and 0.031 g/mL.
