Keragaman Gen TP53INP1 dan Asosiasinya dengan Komposisi Asam Lemak Daging Domba
Abstract
Gen TP53INP1 adalah salah satu gen yang terlibat dalam pengaturan metabolisme lemak dan memiliki keterkaitan dengan komposisi asam lemak pada domba. Penelitian ini bertujuan mengindentifikasi keragaman gen TP53INP1 dan asosiasinya dengan komposisi asam lemak daging domba lokal Indonesia. Total sampel yang digunakan sebanyak 130 sampel DNA ekor domba jantan yang terdiri dari 10 ekor domba barbados cross (DBC), 10 ekor domba compass agrinak (DCA), 20 ekor domba ekor gemuk (DEG), 53 ekor domba ekor tipis (DET), 10 ekor domba komposit garut (DKG), dan 27 ekor domba jonggol (DJG). Identifikasi keragaman gen TP53INP1|NlAIII menggunakan metode Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP). Ukuran produk PCR yang teramplifikasi memiliki panjang basa sebesar 457 bp. Keragaman gen TP53INP1 ditemukan pada semua jenis sampel domba dengan genotipe AA, AG, dan GG. Frekuensi alel TP53INP1|NlAIII berada dalam keseimbangan Hardy-Weinberg kecuali pada domba ekor tipis (DET). Asosisiasi SNP gen TP53INP1 dengan asam lemak menggunakan uji T (P<0,05) pada domba yaitu asam palmitat (C16:0), asam behenat (C22:0), asam lignoserat (C24:0), asam palmitoleat (C16:1), asam elaidat (C18:1n9t), asam gondoat (C20:1), asam nervonat (C24:1), asam oleat (C18:1n9c), dan asam linolelaidat (C18:2n9t). Disimpulkan bahwa SNP gen TP53INP1 mempengaruhi komposisi asam lemak domba. TP53INP1 gene is one of the genes involved in regulating lipid metabolism and is related to the fatty acid composition of sheep. This study aims to identify the diversity of TP53INP1 gene and it’s association with the fatty acid composition of Indonesian local sheeps. The total sample used was 130 ram DNA samples consisting of 10 barbados cross (DBC), 10 agrinak compass sheep (DCA), 20 javanese fat tailed (DEG), 53 javanese thin tailed (DET), 10 garut composite sheep (DKG), and 27 jonggol sheep (DJG). Identification of the diversity of TP53INP1|NlAIII was identified by the Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) method. The amplified PCR product had 457 bp. The diversity of the TP53INP1 gene was found in all types of sheep samples with the genotypes AA, AG, and GG. The allele frequency was in Hardy-Weinberg equilibrium except for the thin tailed sheep (DET). SNP TP53INP1 gene used the T test was associated (P<0,05) with fatty acids in sheeps, such as palmitic acid (C16:0), behenic acid (C22:0), lignoceric acid (C24:0), palmitoleic acid (C16:1), elaidic acid ( C18:1n9t), gondoic acid (C20:1), nervonic acid (C24:1), oleic acid (C18:1n9c), and linolelaidic acid (C18:2n9t). It could be concluded that the SNP TP53INP1 gene effects to the fatty acid composition of lamb.
