Evaluasi Penambahan Ragi Komersial dalam Pakan terhadap Imunitas Ikan Gabus Channa striata yang Diinfeksi Bakteri Aeromonas hydrophila

Ikan gabus merupakan ikan budidaya yang memiliki nilai ekonomis tinggi, salah satunya disebabkan oleh kandungan albumin yang bermanfaat untuk pengobatan pascaoperasi. Budidaya ikan gabus menghadapi kendala infeksi penyakit seperti penyakit motile aeromonad septicaemia (MAS), yang disebabkan oleh infeksi bakteri Aeromonas hydrophila. Bakteri ini menyebabkan kematian hingga 100 %. Salah satu imunostimulan yang potensial digunakan adalah ragi. Penambahan ragi kedalam pakan bermanfaat untuk meningkatkan imunitas ikan. Penelitian ini bertujuan untuk mengevaluasi penambahan ragi komersial dalam pakan untuk meningkatkan imunitas ikan gabus terhadap infeksi bakteri Aeromonas hydrophila.
Peneilitian ini terdiri atas dua tahap. Ikan gabus yang digunakan berasal dari Balai Riset Pemuliaan Ikan (BRPI) yang berukuran 7.37±0,12 cm. tahap satu menggunakan rancangan acak lengkap (RAL) dengan lima perlakuan dan tiga ulangan, masing-masing adalah kontrol (K), penambahan ragi kue 5 g Kg-1 (F5), penambahan ragi tempe 3 g Kg-1 (R3), penambahan ragi tempe 5 g Kg-1 (R5), dan penambahan ragi tempe 7 g Kg-1 (R7). Penelitian tahap dua adalah uji tantang bakteri Aeromonas hydrophila di ikan gabus. Penelitian tahap dua menggunakan rancangan acak lengkap (RAL) dengan enam perlakuan dan tiga ulangan, masing-masing adalah kontrol positif (K+), kontrol negatif (K-), penambahan ragi kue 5 g Kg-1 (F5), penambahan ragi tempe 3 g Kg-1 (R3), penambahan ragi tempe 5 g Kg-1 (R5), dan penambahan ragi tempe 7 g Kg-1 (R7). Parameter yang diambil adalah jumlah konsumsi pakan (JKP), laju pertumbuhan spesifik (LPS), rasio konversi pakan (RKP), dan tingkat kelangsungan hidup (TKH), pola kematian ikan gabus, gejala klinis, parameter gambaran darah, dan ekspresi gen lisozim.
Hasil penelitian tahap satu menunjukkan bahwa JKP berbeda nyata antara R5 (140,12±1,4 g) dengan perlakuan lainnya. LPS tertinggi adalah R3 (5,08±0,12. RKP memiliki nilai 0,70±0,06 – 0,95±0,01. Hasil penelitian tahap dua menunjukkan bahwa perlakuan R3 (56,67 %) dan F5 (50 %) berbeda nyata dengan K+ (13,33 %). Hal ini disebabkan karena kandungan ragi dapat meningkatkan respons imun ikan gabus. Hal ini sesuai dengan total eritrosit ikan gabus pada H+2 perlakuan R3 berbeda nyata antar perlakuan dan memiliki nilai tertinggi yaitu 1,77±0,23 × 106 sel mm-3, hemoglobin 5,8 ± 0,20 g %. Total leukosit dan aktivitas fagositik ikan gabus pada H+4 berbeda nyata antar perlakuan dan memilki nilai tertinggi yaitu 4,97±0,46 × 104 sel mm-3 dan 33,67±3,00 %. Aktivitas lisozim yang memiliki nilai tertinggi adalah F5 (40,00±4,71 IU/ml/menit). Perlakuan R3 terekspresi 1,5 kali dibanding dengan K. Perlakuan R5 terekspresi 4 kali lipat dibanding dengan K. Kesimpulan penelitian ini yaitu Penambahan ragi 3 g Kg-1 dapat meningkatkan imunitas pascauji tantang dengan A. hydrophila. Snakehead is a cultured fish with high economical value, one of which is caused by the albumin content utilized as a post-operation treatment. The snakehead culture confronts a disease infection problem, such as motile aeromonad septicaemia (MAS) caused by Aeromonas hydrophila bacterial infection. These bacteria cause 100 % mortality. The immunostimulant used to prevent the infection of A. hydrophila can be obtained from plant extracts and other chemical compounds as one of which is yeast. Yeast is the mixture of khamir, mold, and bacteria. The addition of yeast in feed can help improve the fish immunity. This study aimed to evaluate the commercial yeast addition in feed for improving the immunity response of the snakehead fish.
This study contained two steps. The first step was a growth performance test. The first step used a completely randomized design with five treatment additions and three replications, namely control (K), 5 g Kg-1 cake yeast (F5), 3 g Kg-1 tempeh yeast (R3), 5 g Kg-1 tempeh yeast (R5), and 7 g Kg-1 tempeh yeast (R7). Parameters obtained were feed intake (FI), specific growth rate (SGR), feed conversion ratio (FCR), and survival rate (SR). The second step was a challenge test with Aeromonas hydrophila bacteria in snakehead fish. The second step used a completely randomized design with six treatments and three replications, namely positive control (K+), negative control (K-), 5 g Kg-1 cake yeast (F5), 3 g Kg-1 tempeh yeast (R3), 5 g Kg-1 tempeh yeast (R5), and 7 g Kg-1 tempeh yeast (R7). Parameters obtained were feed intake (FI), specific growth rate (SGR), feed conversion ratio (FCR), and survival rate (SR), snakehead fish mortality pattern, clinical symptoms, blood profiles, and lysozyme gene expression.
The first step result showed that FI was significantly different among R5 (140,12±1,4 g) and other treatments. The highest SGR rate are R3 (5,08±0,12). Range of RKP are 0,70±0,06 – 0,95±0,01. The second step showed that R3 (56,67 %) and F5 (50 %) were significantly different from K+ (13,33 %). This condition was caused due to the yeast content could enhance the snake-head fish immune response. This followed the snake-head fish total erythrocytes in R3 treatment which was significantly different from other treatments with the highest value of 1,77±0,23 × 106 cells mm-3and hemoglobin of 5,8±0,20 g %. The total leucocytes and phagocity activity of snake-head fish on 4 days after challenge test were insignificantly different among treatments with the highest values of 4,97±0,46 × 104 cells mm-3 and 33,67±3,00%. The highest lysozyme activity was from F5 treatment (40,00±4,71 IU/mL/minutes). The lysozyme gene of R3 treatment was expressed 1.5 times more than K treatment. The lysozyme gene of R5 treatment was expressed 4 times more than K treatment. This study concludes that the addition of 3 g Kg-1 yeast can enhance the fish imunity after challenged with A. hydrophila.
