Diagnosis dan Pengaruh Klon terhadap Epidemi Penyakit Gugur Daun Karet (Hevea brasiliensis Muell. Arg.)

Salah satu penyebab rendahnya produksi karet yaitu adanya gangguan penyakit tanaman. Penyakit gugur daun pada tanaman karet merupakan kendala utama, setelah penyakit jamur akar putih. Saat ini, perkebunan karet di beberapa wilayah Indonesia mengalami kejadian penyakit gugur daun karet baru yang memiliki gejala berbeda dengan penyakit gugur daun yang sebelumnya telah dilaporkan. Penyakit ini juga dilaporkan berkembang luas di beberapa negara Asia Tenggara seperti Malaysia, India, Thailand, dan Sri Lanka. Gejala penyakit berupa bercak daun berbentuk bulat berwarna cokelat dengan pinggiran bercak berwarna lebih gelap. Gejala bercak terdapat pada daun berwarna hijau sampai hijau tua. Daun yang terserang patogen dapat mengalami perubahan warna menjadi kuning atau oranye. Penyakit gugur daun terjadi pada berbagai jenis klon karet dan infeksi berat dapat menyebabkan daun karet menjadi gugur lebih dari 50%. Adanya kejadian penyakit tersebut menyebabkan terjadinya penurunan produksi lateks sampai 45%. Penurunan produksi lateks akibat penyakit menjadi permasalahan penting bagi para petani karet serta pelaku usaha karet.
Banyak dugaan penyebab penyakit, seperti disebabkan oleh cendawan Fusicoccum sp., Colletotrichum spp., dan Pestalotiopsis sp., namun sampai saat ini belum ada diagnosis yang akurat mengenai patogen gugur daun karet dan faktor-faktor yang memengaruhi terjadinya epidemi penyakit pada beberapa klon karet. Oleh karena itu, dilakukan suatu studi untuk mendeterminasi penyebab penyakit gugur daun baru pada karet serta mengkaji pengaruh klon karet terhadap terjadinya epidemi penyakit. Hasil penelitian ini memberikan informasi dalam mengembangkan strategi pengendalian penyakit gugur daun karet yang tepat sasaran, efektif, dan efisien, serta penerapan teknis budidaya yang tepat sebagai upaya pencegahan terjadinya epidemi penyakit.
Diagnosis gugur daun karet dilakukan dengan postulat Koch dan konfirmasi spesies patogen dilakukan melalui identifikasi morfologi dan molekuler. Diagnosis dilakukan terhadap isolat patogen yang berasal dari 4 klon karet (RRIC 100, PB 260, BPM 24, dan GT 1). Isolasi cendawan patogen dilakukan pada media PDA. Inokulasi biakan murni patogen dilakukan pada bibit karet menggunakan metode penempelan miselia cendawan patogen berumur 10 hari. Identifikasi patogen dilakukan berdasarkan karakter koloni dan morfologi, selanjutnya identifikasi molekuler dilaksanakan melalui teknik PCR menggunakan pasangan primer ITS1/ITS4. Dampak klon terhadap terjadinya epidemi dikaji melalui pengamatan kondisi tajuk tanaman menggunakan drone dan persentase keparahan penyakit di lapangan. Selain itu, dilakukan analisis hara tanaman dan data iklim untuk mengetahui hubungannya dengan keparahan penyakit gugur daun di lapangan.
Berdasarkan identifikasi secara morfologi dan molekuler, penyebab gugur daun baru pada tanaman karet yaitu cendawan Pestalotiopsis microspora (Speg.) G. C. Zhao & N. Li. Selanjutnya, penyakit gugur daun karet yang baru disebut penyakit gugur daun Pestalotiopsis. Hasil pengamatan keparahan penyakit menunjukkan bahwa pada awal pengamatan (Januari 2020) keparahan penyakit berkisar antara 13% sampai 25%, namun setiap bulannya mengalami peningkatan hingga pada akhir pengamatan (Juli 2020) keparahan penyakit setiap klon lebih dari 80%. Selanjutnya dilakukan analisis AUDPC dan laju infeksi yang menunjukkan nilai AUDPC berkisar antara 346 unit sampai 505 unit dan nilai laju infeksi berkisar antara 0,54 sampai 1,22. Berdasarkan analisis persentase keparahan penyakit, nilai AUDPC, dan laju infeksi diketahui bahwa kelima klon uji berpengaruh nyata pada epidemi penyakit. Klon RRIC 100, IRR 39, dan PB 260 merupakan klon yang moderat tahan terhadap penyakit gugur daun, sedangkan klon BPM 24 dan GT 1 termasuk klon yang rentan terhadap penyakit gugur daun Pestalotiopsis.
Berdasarkan kajian korelasi unsur kimia tanaman, diketahui bahwa hara tanaman memiliki hubungan korelasi nyata dengan persentase keparahan penyakit. Unsur hara kalium memiliki hubungan korelasi yang sangat kuat terhadap perkembangan penyakit. Unsur hara fosfat dan nitrogen berturut-turut memiliki hubungan korelasi yang kuat dan sedang terhadap perkembangan penyakit gugur daun Pestalotiopsis di lapangan. Analisis cuaca terhadap perkembangan penyakit gugur daun pada 5 klon karet menunjukkan hasil bahwa faktor cuaca mendukung perkembangan penyakit gugur daun di lapangan. Pada saat kelembapan tinggi (87%-89%) dengan curah hujan yang tidak begitu deras (lebih dari 100 mm/hari) dalam waktu yang relatif singkat diikuti dengan periode panas atau tanpa hujan yang panjang maka kondisi tersebut optimal bagi perkembangan penyakit di lapangan. Namun, pada saat hujan deras walaupun kelembapan dan suhu udara tinggi, tidak dapat mendukung perkembangan penyakit di lapangan. Selain itu, faktor kecepatan angin berkisar antara 1,5 km/jam sampai 2,5 km/jam dan lama penyinaran matahari berkisar antara 3 jam/hari sampai 5 jam/hari juga mendukung perkembangan penyakit gugur daun Pestalotiopsis di lapangan.
Hasil penelitian ini memberikan informasi mengenai spesies penyebab penyakit gugur daun baru pada tanaman karet. Hal tersebut menjadi dasar bagi para peneliti untuk mengembangkan penelitian lanjutan terkait karakter bioekologi patogen atau pengembangan strategi pengendalian penyakit. Selain itu, hasil penelitian ini juga memberikan informasi bagi para pelaku usaha karet untuk mencegah terjadinya epidemi penyakit dengan melakukan penanaman karet menggunakan klon yang tahan terhadap penyakit, melakukan pemupukan yang tepat jenis; tepat dosis; tepat waktu; tepat tempat; dan tepat cara, serta melakukan teknis budidaya karet sesuai SOP yang direkomendasikan. One of the causals of low rubber production is the presence of plant diseases. Leaf fall disease in rubber plants is the major disease, after white root disease. Currently, rubber plantations in several areas of Indonesia are experiencing new rubber leaf fall disease which has different symptoms from the previously reported. This disease is also reported to be widespread in several Southeast Asian countries such as Malaysia, India, Thailand, and Sri Lanka. Symptoms of the disease are brown rounded leaf spots with darker fringes. Symptoms of spots on the green or dark green leaves. Leaves that are attacked by pathogens can change color to yellow or orange. The incidence of leaf fall disease occurs in various types of rubber clones and severe infections can cause rubber leaves to fall more than 50%. The occurrence of the disease causes a decrease in latex production up to 45%. The decline in latex production due to disease is an important issue for rubber farmers and rubber industries.
There are many suspicions about the causal agent, such as caused by the fungi Fusicoccum sp., Colletotrichum spp., and Pestalotiopsis sp., but there is no accurate diagnosis of the causal agent and the factors that influence the occurrence of the disease epidemic on several rubber clones. Therefore, a study was carried out to determine the causal agent of new leaf fall disease in rubber plants and the impact of rubber clones on the disease epidemic. The results of this research provide information in developing strategies for controlling leaf fall disease that are precise, effective, and efficient, as well as the application of appropriate cultivation techniques as an effort to prevent disease epidemics.
Diagnosis of leaf fall disease was carried out by Koch's postulates and confirmation of causal agent was carried out by morphological and molecular identification. Diagnosis was carried out by pathogen isolation from 4 rubber clones (RRIC 100, PB 260, BPM 24, and GT 1). Isolation of pathogen was carried out on PDA media. Inoculation of pathogenic cultures was carried out on rubber seedlings using the 10-day old fungal mycelia with attachment method. Pathogen identification was carried out based on the colony and morphological characters, then molecular identification was carried out through PCR techniques using ITS1/ITS4 primer pairs. The impact of clones on the occurrence of disease epidemics was assessed by observing the condition of the plant canopy using drones and the percentage of disease severity in the field. In addition, analysis of plant nutrients and climatic data was carried out to determine the relationship with leaf fall disease severity in the field.
Based on morphological and molecular identification, the causal agent of new leaf fall in rubber plants is the fungus Pestalotiopsis microspora (Speg.) G. C. Zhao & N. Li. Furthermore, a new rubber leaf fall disease is called Pestalotiopsis leaf fall disease. The results of the disease severity observation showed that at the beginning of the observation (January 2020) the disease severity ranged from 13% to 25%, but each month it increased until at the end of the observation (July 2020) the disease severity for each clone was more than 80%. Analysis of AUDPC and infection rates showed that AUDPC values ranged from 346 units to 505 units and infection rates ranged from 0,54 to 1,22. Based on the analysis of the percentage of disease severity, AUDPC value, and infection rate, it was found that the five clones had a significant effect on the disease epidemic. RRIC 100, IRR 39, and PB 260 clones were moderate resistant to leaf fall disease, while BPM 24 and GT 1 clones were susceptible to Pestalotiopsis leaf fall disease.
Based on the study of the correlation of plant chemical elements, it is known that plant nutrients have a significant correlation with the percentage of disease severity. Potassium has a very strong correlation with disease development. Phosphate and nitrogen nutrients have a strong and moderate correlation with the development of Pestalotiopsis leaf disease in the field, respectively. Weather analysis on the development of leaf fall disease in 5 rubber clones showed that weather factors supported the development of leaf fall disease in the field. When the humidity is high (87% -89%) with less heavy rainfall (more than 100 mm/day) in a relatively short time followed by long periods of hot or no rain, these conditions are optimal for disease development in the field. However, during heavy rain, even though the humidity and air temperature are high, it cannot support disease development in the field. In addition, the wind speed factor ranges from 1,5 km/hour to 2,5 km/hour and the sun's exposure time ranges from 3 hours/day to 5 hours/day also supports the development of Pestalotiopsis leaf fall disease in the field.
The results of this research provide information about the species that cause new leaf fall disease in rubber plants. This is the basis for researchers to develop further research related to the bioecological characteristics of pathogens or to develop disease control strategies. In addition, the results of this research also provide information for farmers and rubber industries to prevent disease epidemics by planting rubber using clones that are resistant to disease, carrying out the right type of fertilization; the right dose; the right time; the right place; and the right way, and carry out technical rubber cultivation according to the recommended Standard Operating Procedure.
