      Depolimerisasi Kitosan dan Evaluasi Enkapsulasi Ibuprofen Tersalut Kitosan Bobot Molekul Rendah-Pektin

      Abstract
      Kitosan merupakan polimer alami yang banyak digunakan di bidang medis dan pengantaran obat. Kitosan bobot molekul rendah atau low molecular weight chitosan (LMWC) memiliki kelebihan dibandingkan kitosan bobot molekul tinggi, yaitu solubilitas yang lebih tinggi dalam air sehingga memperluas penerapannya. Dalam penelitian ini, kitosan didepolimerisasi melalui proses hidrolisis dengan HCl. LMWC berhasil diperoleh dengan rendemen rata-rata 89%. Oligomer kitosan dengan bobot molekul terendah, yaitu sekitar 8000 g mol-1 digunakan untuk mengenkapsulasi ibuprofen. Ibuprofen merupakan obat antiinflamasi nonsteroid yang dapat menghasilkan efek samping berupa tukak lambung dan perdarahan pada saluran cerna. Efek samping ibuprofen dapat dikurangi dengan cara enkapsulasi sehingga memperlambat pelepasan obat. Mikrokapsul LMWC-pektin termuat ibuprofen dibuat melalui metode emulsi penguapan pelarut. Kapsul berhasil terbentuk dengan rendemen 90% dan efisiensi sebesar 6.1%. Kapsul yang diperoleh memiliki ukuran yang tidak seragam dan mengalami aglomerasi.
       
      Chitosan is a natural polymer widely used for medical and drug delivery applications. Low molecular weight chitosan (LMWC) has higher solubility in water than the high molecular weight, broadening its applicability. In this study, chitosan was depolymerized by hydrolysis with HCl, yielding an average of 89%. The chitosan oligomer with the lowest molecular weight, approximately 8000 g mol-1, was used to encapsulate ibuprofen. Ibuprofen is a non-steroidal anti-inflammatory drug with adverse effects such as peptic ulcers and gastrointestinal bleeding. The adverse effects of ibuprofen can be overcome by encapsulation to slow down the drug release. The LMWC-pectin microcapsules loaded with ibuprofen were prepared using the emulsion solvent evaporation method giving 90% yield and 6.1% efficiency. The resulted capsules have non-uniform sizes and agglomerated.
