| Abstract: | Nepenthes rafflesiana Jack. merupakan tumbuhan karnivora yang terancam
punah dan memiliki potensi tinggi dalam bioprospeksi serta sebagai tanaman hias.
Penelitian ini mengeksplorasi pengaruh penambahan hormon sitokinin 2-iP pada
konsentrasi kecil terhadap pertumbuhan eksplan N. rafflesiana dalam kultur
jaringan in vitro. Penelitian dilakukan dengan empat variasi konsentrasi 2-iP, yaitu
0 mg/L, 0,001 mg/L, 0,01 mg/L, dan 0,1 mg/L. Pengamatan dilakukan selama 10
minggu terhadap parameter jumlah daun, jumlah tunas, dan tinggi eksplan. Hasil
penelitian menunjukkan bahwa perlakuan dengan konsentrasi 2-iP 0,1 mg/L
memberikan hasil terbaik dalam pertumbuhan elongasi tinggi eksplan, meskipun
secara statistik hanya signifikan pada minggu ke- 2 dan ke-3 dibandingkan
perlakuan lainnya. Penelitian ini menyimpulkan bahwa konsentrasi kecil 2-iP tidak
signifikan berpengaruh terhadap pertumbuhan, dan penggunaan konsentrasi lebih
tinggi direkomendasikan untuk penelitian selanjutnya guna mendapatkan hasil yang
lebih signifikan. Nepenthes rafflesiana Jack. is an endangered carnivorous plant and has high potential in bioprospecting and as an ornamental plant. This study explored the effect of adding cytokinine hormone 2-iP at low concentrations on the growth of N. rafflesiana explants by in vitro tissue culture. The study was conducted with four variations of 2-iP concentrations, namely 0 mg/L, 0.001 mg/L, 0.01 mg/L, and 0.1 mg/L. Observations were made for 10 weeks on the parameters of the number of leaves, number of shoots, and explant height. The results showed that treatment with a concentration of 2-iP 0.1 mg/L gave the best results in the growth of explant height elongation, although statistically significant only in the 2nd and 3rd weeks compared to other treatments. This study concluded that low concentrations of 2-iP did not significantly affect growth, and the use of higher concentrations is recommended for further research to obtain more significant results |
