Abstract: Kumis Kucing (Orthosiphon aristatus) merupakan salah satu dari beberapa tanaman yang memiliki potensi sebagai agen antiobesitas. Penelitian ini bertujuan menguji efektivitas hasil fraksinasi daun kumis kucing sebagai agen antiobesitas berdasarkan kinerjanya dalam menghambat aktivitas lipase pankreas. Sampel diuji kadar airnya, kemudian diekstrak dengan etanol 70% menggunakan metode sonikasi dan dilanjutkan dengan fraksinasi bertingkat menggunakan empat pelarut dengan sifat kepolaran yang berbeda (n-heksana, kloroform, etil asetat, dan akuades). Hasil ekstraksi dan fraksinasi diuji fitokimia (fenolik, flavonoid, dan antioksidan) dan uji inhibisi terhadap lipase pankreas. Hasil penelitian menunjukkan seluruh fraksi dan ekstrak etanol 70% memiliki aktivitas inhibisi bahkan lebih besar dibandingkan dengan kontrol positifnya (orlistat). Fraksi yang memiliki persen inhibisi tertinggi adalah fraksi akuades dengan nilai 94,15%. Kumis kucing berpotensi sebagai agen antiobesitas. Namun perlu dilakukan uji lanjutan secara in vivo untuk membuktikan potensi dari tanaman ini lebih lanjut.
Orthosiphon aristatus is one of the few plants which has the potential as antiobesity agent. This study aimed to measure the effectivity of frationation of its leaves as antiobesity agent based on its performance in inhibiting the activity of pancreatic lipase. Before extraction, the sample was tested for water content. The extraction was performed with sonication method and followed by multilevelled fractionation using four different solvents based on their polarity (n-hexane, chloroform, ethyl acetate, and aquades). Phytochemical (phenolic, flavonoid, and antioxidant) and pancreatic lipase inhibition assays were performed for both crude extract and fractions. The result showed all fractions and crude extract have higher ability to inhibit target enzyme than the control positive (orlistat). The inhibition activity of aquades fraction was 94,15% which was the highest value out of all the sample tested. As the results had been proven, the potential of Orthoshiphon aristatus as antiobesity must be investigated through in vivo assay in order to prove the effectivity.
