Sensor Asam Urat Berbasis Elektrode Karbon Kaca Termodifikasi Komposit Grafena Oksida/Emas Nanorod

Asam urat merupakan produk metabolisme purin yang terdapat dalam cairan fisiologis tubuh. Kadar asam urat yang tinggi mengakibatkan terjadinya penumpukan kristal asam urat yang dapat memicu penyakit hiperurisemia, asam urat akut, hingga batu ginjal. Teknik deteksi asam urat yang sensitif sangat diperlukan untuk pemantauan kadar asam urat dan mencegah penyakit yang ditimbulkannya. Teknik deteksi yang biasa digunakan untuk mengukur kadar asam urat di laboratorium uji ialah kolorimetri dan spektrofotometri. Teknik tersebut memiliki sensitivitas yang baik, namun hanya dapat dilakukan di laboratorium uji oleh analis yang terlatih. Di sisi lain, metode elektrokimia dapat mendeteksi asam urat berdasarkan reaksi oksidasi asam urat pada permukaan elektrode.
Pada penelitian ini, suatu sensor elektrokimia untuk deteksi asam urat dikembangkan berbasis elektrode karbon kaca (GCE) termodifikasi material konduktif berupa komposit grafena oksida (GO) dan emas nanorod (AuNR). Metode Hummers termodifikasi digunakan untuk menyintesis GO. Adapun AuNR disintesis menggunakan metode termediasi benih dengan jumlah AgNO3 yang bervariasi (0,4; 0,5; dan 0,6 mL) untuk menghasilkan rasio aspek yang berbeda. GO serta AuNR dengan rasio aspek 3,3; 3,4; dan 3,5; berhasil disintesis. Hal ini dibuktikan dengan hasil karakterisasi menggunakan instrumen spektroskopi Raman, FTIR, UV-Vis-NIR, XRD dan TEM. Kedua bahan tersebut dicampur menjadi komposit pemodifikasi untuk mengembangkan GCE/GO/AuNR dengan berbagai aspek rasio AuNR. Selanjutnya kinerja elektrokimia GCE/GO/AuNR diselidiki menggunakan teknik voltammetri pulsa diferensial (DPV).
Penggunaan aspek rasio AuNR yang berbeda menghasilkan respon elektrokimia berbeda pada pengukuran DPV asam urat. Komposit GO/AuNR rasio 3,5 menunjukkan arus puncak yang lebih tinggi dibandingkan aspek rasio lainnya sehingga digunakan untuk tahap selanjutnya. Rancangan percobaan central composite design (CCD) selanjutnya diterapkan untuk mendapatkan komposisi GO/AuNR-3,5 optimum, yaitu yang mampu memberikan arus puncak oksidasi asam urat maksimum. GCE/GO/AuNR-3,5 pada komposisi optimum memiliki rentang linear pengukuran asam urat pada konsentrasi 10−100 μM (R2 = 0,9917), batas deteksi dan batas kuantisasi masing-masing sebesar 0,4 μM dan 1,0 μM. Selanjutnya, sensor yang diusulkan menunjukkan stabilitas, reprodusibiltas, selektivitas (dengan keberadaan interferen asam askorbat, dopamin, glukosa, urea, magnesium), dan berhasil digunakan untuk menentukan kandungan asam urat dalam sampel urin manusia dengan metode adisi standar. Temuan pada penelitian ini menunjukkan komposit GO/AuNR merupakan material yang menjanjikan untuk pengembangan sensor asam urat yang sensitif. Uric acid is a product of purine metabolism found in the body's physiological fluids. High uric acid levels resulted in urate crystals that can trigger hyperuricemia, gout, and kidney stones. The uric acid detection technique become important to monitor uric acid levels and prevent the disease it caused. Common detection techniques used to measure uric acid levels in test laboratories are colorimetry and spectrophotometry. The technique has good sensitivity but can only be performed in a test laboratory by a qualified analyst. On the other hand, electrochemical methods can detect uric acid based on the uric acid oxidation reaction on the electrode surface.
In this study, an electrochemical sensor for detecting uric acid was developed based on a modified glass carbon electrode (GCE) graphene oxide (GO) and gold nanorod (AuNR) composites. The hummers method was used to synthesize GO. The AuNR was synthesized using the seed-mediated method with varying amounts of AgNO3 (0.4; 0.5; and 0.6 mL) to produce different aspect ratios. GO and AuNR with an aspect ratio of 3.3; 3.4; and 3.5; successfully synthesized and characterized using the instruments of Raman spectroscopy, FTIR spectroscopy, UV-Vis-NIR spectroscopy, XRD and TEM. The two materials were mixed as composite as modifier to develop GCE/GO/AuNR with various AuNR aspect ratios. Furthermore, the electrochemical performance of GCE/GO/AuNR was measured using the differential pulse voltammetry (DPV) technique.
The use of different AuNR aspect ratios resulted in different DPV electrochemical responses in the measurement of uric acid. The GO/AuNR composite ratio of 3.5 showed a higher peak current than the other aspect ratios, so it was chosen for the next method step. The central composite design (CCD) experimental design was then applied to obtain the optimum composition of GO/AuNR-3.5, which was able to provide the maximum peak current of uric acid oxidation. GCE/GO/AuNR-3.5 at the optimum composition has a linear measurement range of uric acid at a concentration of 10−100 μM (R2 = 0.9917), the detection limit and quantization limit are 0.4 μM and 1.0 μM, respectively. Furthermore, the proposed sensor demonstrated stability, reproducibility, selectivity (in the presence of interferences of ascorbic acid, dopamine, glucose, urea, and magnesium), and was successfully used to determine uric acid in human urine by the standard addition method. The findings in this study indicate GO/AuNR composites are a promising material for the development of sensitive uric acid sensors.
