Penggunaan rhizobakteri tahan masam seperti /lradyrhlzobium japonicum merupakan salah satu upaya untuk meningkatkan produksi kedelai di tanah masam. IL. japonlcum dapat bersimbiosis dengan tanaman kedelai dan menambat nitrogen bebas. Penelitian ini bertujuan untuk mengukur pertumbuhan dan produksi kedelai varietas Wilis yang diinokulasi dengan ll.. japonicum dan pemberian pupukdi tanah masam. Penelitian ini menggunakan bakteri ll.. jaeonicum BJ 11 (19) has ii mutagenesis transposon dan BJ 11 (wt) tipe liar yang toleran rerhadap asam. Perlakuan yang diberikan pada kedelaz wlah pemberian inokulan BJ 11 (19), BJ 11 (wt) yang masing-masing dikombinasilcan dengan pemberian kompos dan pupuk N (50 dan 1000/o dos is). Percobaan Japang dila/cukan di Cikabayan, Darmaga, Bogar menggunalcan rancangan kelompok lengkap teracak dengan 15 perlalcuan dan 3 ulangan. Hasil pengamatan memmjukkan bahwa pemberian inokulan "fl.. iaponicum toleran asam BJ 11 (wt), kompos dan pupuk N (J 0 g m·1) secara bersama-sama dapat meningkatkan tinggi tanaman, bobot kering akar don lajuk, j11mlah nodu/, bobot kering nodul, aktivilas nitrogenase, jumlah polong dan biji per tanaman maupun per satuan luas, bobot biji per tanaman maupun per satuan luas, kandungan N biji yang ditanam di tanah masam pH 4. 7.

Melastoma malabathricum L. grows well in acid soil with high level a/soluble aluminum. One of the important proteins in the detoxifying acid and aluminum stress is a plasma membrane H' -ATPase protein encoded by PMA gene. The objective of Ihis research was 10 isolale and clone Ihe cDNA fragmenl of MmPMA encoding plasma membrane H~ -ATPase from M. malabalhricum L. By reverse Iranscriplion. 10101 cDNA had been synthesized from Ihe 10101 RNA as lemplate. The fragmenl of MmPMA cDNA had been successfully isolaled by PCR by using 10101 cDNA as lemplale and PMA primer designed from conserved region Jor corresponding gene. This fragment had been success/ully inserted into pGEM-T Easy and the recombinant plasmid was sl/cces!J!ully introduced into ft.. coli DH5a. Nucleotide sequ.ence analysis showed that the length of MmPMA frogmen I is 806 bp encoding 268 amino acids. Local alignment analysis based on nucleotide of mRNA showed Ihal MmPMA fragmenl was 81% idenlical 10 pari of PMA of Sesbania roslrala Juglam regia and Prunus persico. Based on deduced amino acid sequence, MmPMA was 94% idenlicallo pari of PMA of Juglam regia; 93% 10 PMA ofS. roslrala, and Arabidopsis Iltaliana. MmPMAfragmenl has pltosphO/ylalion inlermediale domain (DKTGT) and ATP binding domain (KGAp' DPPR. MITGD, andGDGVN).

Extension of jatropha planting on coastal sandy soil is an effort to look for an alternatif area to other soil types, this experiment is one of a series experiment aiming to classify jatropha based on their drought resistance, and to determine watering interval causing drought stress. The experiment was arranged using Split Plot Design with watering interval (once in 1, 3, 5, 7, and 9 day (s)) as main plot, and jatropha genotypes (IP-1A, IP-1M, IP-1P, IP-2A, IP-2M, IP-2P, Unggul Lokal (NTB), Gundul and Daun Kuning) as sub plot. The seeds of genotypes were planted in black polybag with coastal sandy soil media. The parameters observed were plant height, stem diameter, number of leaf, leaf width and plant dry weight, and the data collected were analyzed using Analysis of Variance, DMRT, Regrression and Cluster Analysis. The result showed that IP-1A, IP-2M, Gundul and IP-1M are the resistant genotypes, whereas Unggul Lokal, Daun Kuning, IP-2A and IP-2P are the susceptible genotypes. Once in one and three day (s) are optimum watering intervals, and once in nine days is a watering interval causing drought stress.

A study was conducted to determine Jatropha fruits ripening uniformity on its tree and bunches characteristics. To get more information about its ripening off, the tree characteristics a respiration pattern was determined. The data were collected on fi ve trees for each ten selected accessions (n=50) for fruit ripening uniformity on its trees study and fi fteen bunches for each seven selected accessions (n=105) for fruits bunches ripening uniformity study at two jatropha pilot projects at Sabah state of Malaysia at the end of March 2009 when the trees were exactly one year old. This study confi rmed that heterogeneous ripening occured in all jatropha accessions and within individual jatropha bunches. Respiration tests confi rmed that jatropha is a climacteric fruit. The results showed an upsurge in CO2 production at the end of ripening and at the beginning of senescence. This study has revealed indications of the possibility to harvest jatropha fruit in bunches rather than harvesting individual ripe fruit, which could potentially improve harvest effi ciency by reducing harvesting time and drudgery.
