To increase variant of Coleus amboinicus Lour. can do by mutation induction using ethyl methane sulphonate. Some studies report that the EMS successful to increase variability genetic and produce mutan plants. The objectives of this study were 1) to determine the interaction of treatments in MV1, MV2 and MV3, 2) to obtain some mutants until MV3, and 3) to determine the heritability estimates value in MV2 and MV3. The experiment was arranged in Randomized Complete Block Design (RCBD) with three replication. The materials used Coleus amboinicus Lour. cuttings shoots. The concentration of EMS consists of 0.00, 0.50, 0.75, 1.00, and 1.25% with application soak method (100 minutes) and drops (3 drops pipette). The results showed that interaction between the method of application and concentration of EMS were only found in plant height and number of leaf (MV1). Some mutants were found which have different morphological appearance to the control. The mutants were R1.25,1 (MV1-3), R0.5,9 (MV2-3), T1.25,6 (MV2-3), and T1.00,6 (MV3). The heritability estimates were moderate for leaf length and leaf width in MV3.id
