This study aims to determine the rutin production of buckwheat seed on different altitude and planting distance. This research carried out in the Kopo garden Bogor and IPB experimental garden Pasir Sarongge Cianjur, West Java, from May to July 2012. The method used was complete random design with factorial of two factors that is altitude and planting distance with three repetition. The first factor is the altitude
consists of two levels, that is an altitude of 600 meters above sea level (m asl) and an altitude of 1150 m asl, and the second factor is planting distance consists of to levels: the planting distance of 2.5 cm x 20 cm and 10 cm x 20 cm. Analysis of rutin levels using HPLC method. Variables measured were the grain weight, the rutin content, and rutin production of buckwheat seed. The results showed the differences in altitude and planting distance gave significant effect (P<0.05) to the grain weight. Altitude difference
gave highly significant effect (P<0.01) to the rutin content, while interactions between planting distance and altitude influence is not significant (P>0.05) to the rutin content. Differences in altitude, planting distances and interactions between altitude and planting distances gave significant influence (P<0.01) in the rutin production of buckwheat seed.
