      Abstract
      This research was conducted to study characteristics (motility, viability, plasma membrane integrity) and intact acrosome status of ram’s spermatozoa cryopreserved in tris-egg yolk or tris-skim milk extenders with one or two step method. Semen was collected from local ram, devided and extended in tris-egg yolk or tris-skim milk by using one step method (OS-KT and OS-SK) and two step method (TS-KT and TS-SK). The results showed that equilibrated spermatozoa had similar quality in all groups (P>0.05). Percentage of post-tahwed motility spermatozoa in OS-SK group was lower than OS-KT group (P<0.05). Percentage of spermatozoa viability, plasma membrane integrity and intact acrosome status were not significantly different among groups (P>0.05). In conclusion, tris-egg yolk extender was better than tris-skim milk extender to prevented the motility of spermatozoa after cryopreservation. Futhermore, one or two step methods were not significantly effected to the quality of post-thawing spermatozoa.
