dc.description.abstractThis study was aimed to find the best method of regeneration through somatic embryogenesis Mandarin var Batu 55 best as results of in vitro poliploidization by colchicine. This study consists of: the proliferation of callus, maturation of somatic embryo, germination of somatic embryos, elongation of roots and shoots. The best concentration of phytagel for callus proliferation was 2.5 g/l with MS modified Vitamin MW, and 500 mg/l of malt extract. Maturation of somatic embryos with ABA concentration of 0.5 mg/l on MS basic medium, vitamins MW, 100 mg/l malt extract was the best medium to produce cotyledons. MS modified by the addition of vitamin MS + 2.5 mg/l GA3 + 500 mg/l malt extract was the best medium for the formation of plantlets with a percentage of 43%. Elongation of roots and shoots can use MS medium with vitamins MS + IBA 0.5 mg/l + 500 mg/l malt extract to get the best results.en
