      Penentuan kestabilan dan linearitas pada biosensor asam urat menggunakan urikase dari lactobacillus plantarum termodifikasi zeolit secara elektrokimia

      Abstract
      Penggunaan biosensor untuk mengukur asam urat telah banyak dikembangkan dengan menggunakan urikase yang bekerja spesifik pada asam urat. Urikase murni sangat mahal harganya, sebagai alternatif digunakan urikase dari Lactobacillus plantarum untuk pengukuran asam urat. Namun, urikase yang dihasilkan dari L.plantarum memiliki stabilitas serta linearitas yang masih rendah. Oleh karena itu, pada penelitian kali ini enzim urikase L.plantarumdiimobilisasi pada zeolit alam (Bayah, Banten) sehingga meningkatkan stabilitas dan linearitasnya. Stabilitas dari biosensor asam urat bertahan hingga 22 hari. Pada rentang konsentrasi 0.4-4 mM dihasilkan sensitivitas sebesar 3.47 μA mM-1 dengan R2= 97.57%. Kondisi optimum yang diperoleh urikase L.plantarumyang diimobilisasi pada zeolit alam dengan desain central composite adalah pH 10, suhu 40 oC, massa zeolit 255 mg, dan konsentrasi asam urat 3.8 mM.
       
      Biosensor for uric acid measurement has been developed using uricase which works specifically for uric acid. uricase is expensive, therefore uricase from Lactobacillus plantarum was used for uric acid measurement as an alternative.However, the uricase from L.plantarumhaslow stability and linearity. Therefore, in this study, uricase from L.plantarum was immobilized on natural zeolite (Bayah, Banten) in order to increase its stability and linearity. Stability of uric acid biosensor could sustained up to 22 days. Sensitivity of 3.47 μA mM- 1 was obtained at concentration range of 0.4−4 mM with R2=97.57%. The optimum condition for the immobilized uricase with central composite design was at pH 10, temperature of 40 oC, uric acid concentration of 3.8 mM, and zeolite mass of 255 mg.
