      Storability Vigor Detection Using Accelerated Aging Method on Upland Rice (Oryza sativa L.) Seeds

      Abstract
      Storability is an important seed character indicating how long the seed could maintain the viability. Accelerated aging with physical or chemical treatments are methods for detecting seed vigor especially storability (storability vigor, VDS). The aims of the research were: 1) to find out a simple accelerated aging method suitable for testing storability vigor of upland rice seeds, 2) to identify upland rice genotypes with high storability vigor. Experiments were conducted in the Seed Science and Technology Laboratory, Plant Analysis and Chromatography Laboratory, Soil Chemical Laboratory, Bogor Agricultural University, from January 2011 to October 2011. Three methods of accelerated aging, 45oC and 100% RH, 96% ethanol vapour, and soaking in 96% liquid ethanol, were applied to seeds of 5 rice varieties i.e. Situ Patenggang, Limboto, Inpago 4, Inpago 5, Inpago 6. The most suitable method was used to evaluate the storability vigor of 26 upland rice genotype seeds. Result of the experiment showed that accelerated aging by soaking seed in liquid ethanol 96% for 3 minutes and 24 seconds decreased the average germination percentage from 94.67% to 84.13% and seedling growth rate from 17.97%/etmal to 15.11%/etmal. However, biochemical analysis showed that there were no significant differences between the treated and untreated seed for electroconductivity, leakage of protein, sugar and phosphorous variables. The ethanol treatment might disturb fungtional proteins including enzymes but not the cell membrane. Evaluation of 26 upland rice genotypes showed that breeding lines B12154D-MR-10, B1215D-MR-11, and B11602-MR- 1-52 performed high storability vigor.
