A study to investigate the rate of head-to-head sperm agglutination and the distribution of antiagglutinin in plasma collected fmm epihdymis and ejaculate was carried out in an effort to optimize their use for in vitro fertilization. The rate of agglutination and the distribution of antiagglutinin were determined by counting the number ofhead-to-head sperm agglutination &r 1,3,5 and 7 hour incubations in Krebs Ringer-HEPES media containing plasma collected from epididymis and ejaculate. The rate of head-to-head sperm agglutination decreased according to the times of incubations. The rate of agglutination was significantly (Pc0.05)lo wer on the sperm collected from cauda epididymal than those collected from caput, corpus epididymal and ejaculate. The result also indicated that the distribution of anti-agglutinin be higher in cauda epididymis than that in other areas. The reduction of agglutination was also obselved on sperm collected from cauda and ejaculate cultured in vitro in KR-HEPES containing the plasma of cauda epididymal. A further investigation is however necessary to confirm whether the antiagglutinin of plasma from cauda epididymis can be used in the sperm preparation for in uitro fertilization. Key words: sperm, agglutination, antiagglutinin, epididymis, plasma
