The application of root nodule bacteria in field faced constraint of inconsistency of its effectivity. The experiment was conducted to characterize 43 superior strains of Bradyrhizobium japonicum and Sinorhizobium fredii from Indonesia based on schizotyping by Pulsed-Field Gel Electrophoresis (PFGE). Schizotyping requires a technique to isolate an intact genomic DNA, the rare-cutting restriction endonucleases, appropriate molecular size markers, and electrophoresis unit. Results indicated that this technique could separate DNA fragments of soybean root nodule bacteria efficiently which had a similarity of physiological and morphological characteristics. By using this technique, 23 strains of B. japonicum consisted of 19 Spel-schizotype and 20 strains of S. fredii comprised of 13 Spel-schizotype.
