      Identifikasi Thrips parvispinus Karny (Thysanoptera: Thripidae) Asal Dua Lokasi di Indonesia Berdasarkan Gen mtCOI

      Abstract
      Thrips parvispinus merupakan salah satu hama invasif yang menyerang banyak jenis tanaman. Serangga ini berukuran kecil sehingga identifikasi secara morfologi terkadang mengalami kesulitan. Penelitian ini bertujuan mengidentifikasi thrips dan mendeteksi variasi haplotipe T. parvispinus berdasarkan gen mithocondrial cytochrome oxidase subunit I (mtCOI). Serangga dikoleksi dari Lebak (Banten) dan Cirebon (Jawa Barat). Serangga diekstraksi menggunakan metode gabungan (bulk extraction) dari 25 individu per lokasi sebagai strategi diagnostik cepat, lalu diamplifikasi dengan primer LCO1490 dan HCO2198 (~700bp). Hasil analisis urutan basa menunjukkan sekuens Cirebon memiliki homologi 100% dengan sekuens global, sedangkan sekuens Lebak menunjukkan variasi minor dengan identitas 99,06-99,22%. Analisis filogenetik menempatkan kedua sekuens dalam satu klade monofiletik yang kokoh dengan sekuens global. Tingginya homologi (>99%) mengindikasikan bahwa sekuens dari kedua lokasi didominasi oleh satu garis keturunan yang sama. Penelitian ini memvalidasi bahwa penggunaan gen mtCOI mampu digunakan untuk identifikasi molekuler cepat T. parvispinus di Indonesia.
       
      Thrips parvispinus is an invasive pest that infests various plant species. Due to its minute size, morphological identification can be challenging. This study aims to identify thrips and detect haplotype variations of T. parvispinus based on the mitochondrial cytochrome oxidase subunit I (mtCOI) gene. Insects were collected from Lebak (Banten) and Cirebon (West Java). DNA extraction was performed using a bulk extraction method on 25 individuals per location as a rapid diagnostic strategy, followed by amplification using primers LCO1490 and HCO2198 (~700 bp). Nucleotide sequencing results showed that the Cirebon sequence shared 100% homology with global sequences, whereas the Lebak sequence exhibited minor variations with 99.06–99.22% identity. Phylogenetic analysis placed both sequences within a robust monophyletic clade alongside global sekuenses. The high degree of homology (>99%) indicates that populations in both locations are dominated by a single shared lineage. This study validates that the mtCOI gene is effective for the rapid molecular identification of T. parvispinus in Indonesia.
