      Isolasi dan Karakterisasi Bakteri dari Kawasan Ekowisata Mangrove Pantai Indah Kapuk yang Berpotensi sebagai Metanotrof

      Abstract
      Emisi gas rumah kaca, terutama metana (CH4), menjadi isu global karena kontribusinya terhadap pemanasan global. Metana memiliki kemampuan pemanasan 28 kali lebih kuat daripada karbon dioksida. Lingkungan mangrove, termasuk kawasan buatan seperti Kawasan Ekowisata Mangrove Pantai Indah Kapuk, Jakarta Utara, merupakan penghasil metana yang signifikan sekaligus menjadi habitat bakteri metanotrof. Bakteri ini adalah penyerap CH4 biologis utama di atmosfer, menggunakan enzim Metana Monooksigenase (MMO) untuk mengoksidasi metana sebagai sumber karbon dan energi. Penelitian ini bertujuan untuk mengisolasi dan mengkarakterisasi bakteri dari kawasan tersebut yang berpotensi sebagai metanotrof. Metode yang digunakan di antaranya pengambilan sampel, isolasi dan pemurnian bakteri metanotrof, karakterisasi makroskopis bakteri, uji kemampuan pertumbuhan bakteri dalam media dengan sumber karbon sukrosa dan glukosa, pewarnaan Gram bakteri, dan uji katalase. Sebanyak tujuh isolat berhasil diperoleh dan menunjukkan variasi karakter morfologi koloni serta pertumbuhan. Hasil karakterisasi menunjukkan ketujuh isolat merupakan bakteri Gram negatif dengan bentuk sel basil dan menunjukkan hasil positif pada uji katalase. Semua isolat, termasuk kontrol positif Methylocystis parvus, menunjukkan pertumbuhan optimal pada media NMS dengan metanol sebagai sumber karbon, yang mengindikasikan potensi mereka sebagai metanotrof. Namun, isolat KEM2, KEM5, dan KEM6 juga menunjukkan kemampuan tumbuh pada media dengan sukrosa dan glukosa, menunjukkan potensi fleksibilitas metabolik.
       
      Greenhouse gas emissions, especially methane (CH4), have become a global issue due to their contribution to global warming. Methane has a warming potential 28 times higher than carbon dioxide. Mangrove environments, including artificial areas such as the Pantai Indah Kapuk Mangrove Ecotourism Area, North Jakarta, are significant methane producers and habitats for methanotrophic bacteria. These bacteria are the main biological CH4 sinks in the atmosphere, using the enzyme methane monooxygenase (MMO) to oxidize methane as a carbon and energy source. This study aimed to isolate and characterize bacteria from the area with potential as methanotrophs. Methods used included sampling, isolation and purification of methanotrophic bacteria, macroscopic characterization, growth tests on media with sucrose and glucose as carbon sources, Gram staining, and catalase tests. Seven isolates were obtained, showing variation in colony morphology and growth. Characterization results indicated that all isolates were Gram-negative rods and catalase-positive. All isolates, including the positive control Methylocystis parvus, showed optimal growth on NMS medium with methanol as the carbon source, indicating their potential as methanotrophs. However, isolates KEM2, KEM5, and KEM6 also grew on media with sucrose and glucose, indicating metabolic flexibility.
