      Genetic Diversity of Gamma-Irradiated Potato (Solanum tuberosum L.) Clones of cv. IPB PAU1

      Abstract
      The potato cultivar IPB PAU1 has been developed by the Biotechnology Center of IPB, but its genetic characteristics still require improvement through mutagenesis. Several clones of IPB PAU1 have been generated through gamma rays irradiation at doses 20 Gy and 30 Gy. However, the molecular profiles of these gamma-irradiated clones have not yet been characterized. This study aimed to analyze the genetic diversity between the IPB PAU1 potato cultivar and its gamma-irradiated clones using molecular markers Random Amplified Polymorphic DNA (RAPD) and Inter-Simple Sequence Repeat (ISSR). DNA was isolated using the Cetyl-Trimethyl Ammonium Bromide (CTAB) method, then amplified with eight RAPD primers and three ISSR primers. The results showed a polymorphism level of 38.92% for RAPD and 55.55% for ISSR. Clustering and Principal Component Analysis (PCA) showed genetic separation, especially in clone 30.9. These findings indicate that gamma rays irradiated induced mutation is effective in generating mutant clones with diverse genetic for early selection of superior varieties.
       
      Kentang kultivar IPB PAU1 telah dikembangkan oleh Pusat Bioteknologi IPB, tetapi karakter genetiknya masih memerlukan perbaikan melalui mutagenesis. Beberapa klon kentang kultivar IPB PAU1 telah dihasilkan melalui iradiasi sinar gamma dengan dosis 20 Gy dan 30 Gy. Namun, profil molekuler klon-klon hasil iradiasi tersebut belum dikarakterisasi. Penelitian ini bertujuan menganalisis keragaman genetik antara kentang kultivar IPB PAU1 dan klon-klon hasil iradiasinya menggunakan marka molekuler Random Amplified Polymorphic DNA (RAPD) dan Inter-Simple Sequence Repeat (ISSR). DNA diisolasi menggunakan metode Cetyl-Trimethyl Ammonium Bromide (CTAB), lalu diamplifikasi dengan delapan primer RAPD dan tiga primer ISSR. Hasil analisis menunjukkan tingkat polimorfisme sebesar 38,92% untuk RAPD dan 55,55% untuk ISSR. Analisis pengelompokan dan analisis komponen utama (PCA) menunjukkan pemisahan genetik, terutama pada klon 30.9. Temuan ini mengindikasikan bahwa mutasi induksi melalui iradiasi sinar gamma efektif dalam menciptakan klon mutan dengan genetik yang beragam untuk seleksi awal varietas unggul.
