      Pengujian Toksisitas Ekstrak Daun Bakau Hitam (Rhizophora mucronata) sebagai Bahan Baku Obat

      Abstract
      Rhizophora mucronata merupakan salah satu jenis bakau yang ada di Indonesia. Daun bakau hitam (Rhizophora mucronata) sering dijadikan sebagai obat. Penelitian ini bertujuan menentukan toksisitas ekstrak daun bakau hitam (Rinophora mucronata) terhadap Artemia salina dan sel Vero serta kandungan komponen aktif dari ekstrak daun bakau hitam. Ekstraksi dilakukan dengan metode muserasi. Perlakuan pada penelitan ini adalah jenis pelarut untuk maserasi yaitu metanol, etil asetat dan n-heksana. Rancangan percobaan pada pengujian toksisitas pada Artemia salina menggunakan metode Rancangan Acak Lengkap dengan uji lanjut Duncan. Pengujian toksisitas ekstrak terhadap sel Vero dilakukan dengan metode Kancangan Acak Lengkap Faktorial dengan uji lanjut Duncan. Hasil pengujian toksisitas terhadap Artemia salina menunjukkan ekstrak n-heksana bersifat toksik rendah dengan nilai LC50 294,96 ppm dan ekstrak metanol bersifat tidak toksik dengan nilai LC50 1425,37 ppm. Hasil pengujian toksisitas terhadap sel Vero menunjukkan ekstrak metanol dan ekstrak etil asetat bersifat tidak toksik. Ekstrak n-heksana bersifat toksik karena memiliki nilai viabilitas kurang dari 50% pada konsentrasi kurang dari 30 ppm.
       
      Rhizophora mucronata is one of mangroves in Indonesia. The black mangrove (Rhizophora mucronata) leaf is often used as a medicine. This study aims to determine the toxicity of black mangrove (Rhizophora mucronata) leaf extract on Artemia salina and Vero cells and the active component of black mangrove leaf extract. Extraction used maceration method. The treatment of this research was the solvent used for extraction including methanol, ethyl acetate and n-hexane. The experimental design on the toxicity test of Artemia salina was performed using the Completely Randomized Design Method with Duncan Test. Examination on toxicity of extract on Vero cell viability was done by Factorial Random Design Complete Design with Duncan Test. Toxicity test results of Artemia salina showed n-hexane extract low toxic with LC50 294,96 ppm and methanol extract not toxic with LC50 1425,37 ppm. The results of the toxicity test on Vero cells showed that methanol extract and ethyl acetate extract were non-toxic. The n-hexane extract was town because it has a value of less than 50% viability at a concentration of less than 30 ppm.
