      Identifikasi Spesies Gastropoda di Pulau Pari, Jakarta Berdasarkan Morfologi dan DNA Barcoding Gen 12S

      Abstract
      Ekosistem Pulau Pari menghadapi tekanan ekologis yang berdampak pada kelimpahan gastropoda. Penelitian ini bertujuan mengidentifikasi spesies gastropoda berdasarkan morfologi dan DNA barcoding gen mitokondria 12S. Sebanyak 56 individu dikoleksi dari empat stasiun di Pulau Pari dan dianalisis melalui pengukuran morfometrik dan sekuensing genetik. Analisis morfologi mengungkap lima spesies dari tiga famili, dengan Terebralia sulcata sebagai spesies dominan. Analisis DNA menggunakan polymerase chain reaction, elektroforesis, dan BLAST NCBI mengidentifikasi tujuh spesies dengan keberhasilan identifikasi tingkat spesies sebesar 32,1%. Rekonstruksi pohon filogeni menunjukkan penempatan spesies yang sesuai pada klade taksonomi masing-masing, dan analisis Automatic Barcode Gap Discovery (ABGD) mengelompokkan sampel ke dalam enam grup spesies. Temuan penting dari studi ini menunjukkan keberadaan spesies kriptik pada Littoraria intermedia dan mendukung efektivitas gen 12S sebagai penanda molekuler alternatif dalam identifikasi gastropoda bercangkang. Penelitian ini memberikan kontribusi data genetik dan morfologi untuk mendukung konservasi serta manajemen keanekaragaman hayati di kawasan ekosistem Pulau Pari.
       
      The ecosystem in Pari Island faces ecological pressure that may affect gastropod abundance. This study aims to identify gastropod species based on morphological characteristics and mitochondrial 12S gene DNA barcoding. A total of 56 individuals were collected from four stations and analyzed using morphometric measurements and genetic sequencing. Morphological analysis revealed five species from three families, with Terebralia sulcata as the dominant species. DNA analysis involved polymerase chain reaction, electrophoresis, and NCBI BLAST, resulting in seven species identified and a 32.1% success rate at species level. Phylogenetic tree reconstruction confirmed accurate taxonomic placement, and Automatic Barcode Gap Discovery (ABGD) analysis grouped samples into six species clusters. A key finding was the presence of cryptic species in Littoraria intermedia, supporting the effectiveness of the 12S gene as an alternative molecular marker for shelled gastropod identification. This study contributes genetic and morphological data to support biodiversity management and conservation efforts in Pari Island ecosystems.
