      Kandungan Prolin dan Asam Askorbat Hopea sangal (Cengal) pada Cekaman Kekeringan

      Abstract
      Hopea sangal (Dipterocarpaceae) merupakan spesies langka dan terancam punah yang ditemukan kembali di Kabupaten Pasuruan pada 2014. Famili ini adaptif terhadap kekeringan melalui akumulasi prolin dan asam askorbat (ASA), namun respons fisiologis H. sangal terhadap cekaman tersebut belum diketahui. Penelitian ini bertujuan memahami kemampuan adaptasi H. sangal terhadap cekaman kekeringan melalui analisis kandungan prolin dan ASA sebagai indikator fisiologis ketahanan terhadap kekeringan. Perlakuan meliputi kontrol, kekeringan (tanpa penyiraman selama 3, 6, dan 9 hari), serta pemulihan (penyiraman pada hari ke-10 dan pengamatan pada hari ke-11). Parameter yang diamati meliputi kandungan prolin, ASA, morfologi tanaman yang mencakup tinggi batang, jumlah dan gugurnya daun, jumlah cabang, serta faktor lingkungan seperti suhu, intensitas cahaya, dan kelembapan. Hasil menunjukkan bahwa akumulasi prolin meningkat secara signifikan hingga 6 hari setelah perlakuan (HSP), kemudian menurun selama pemulihan. Kandungan ASA stabil selama kekeringan dan meningkat setelah penyiraman. Tinggi batang dan jumlah daun meningkat, sedangkan jumlah cabang tidak menunjukkan perbedaan yang signifikan.
       
      Hopea sangal (Dipterocarpaceae) is a rare and endangered species rediscovered in Pasuruan Regency in 2014. This family is adaptive to drought through proline and ascorbic acid (ASA) accumulation, but the physiological response of H. sangal to such stress remains unknown. This study aims to understand the adaptive capacity of H. sangal to drought stress through the analysis of proline and ASA contents as physiological indicators of drought tolerance. Treatments consisted of control, drought (no watering for 3, 6, and 9 days), and recovery (watering on day 10 and observation on day 11). Observed parameters included proline and ASA content; plant morphology, including stem height, number and abscission of leaves, number of branches; as well as environmental factors such as temperature, light intensity, and humidity. Results showed that proline accumulation significantly increased until the sixth day after treatment (DAT), then decreased during recovery. ASA content remained stable during drought and increased after watering. Stem height and number of leaves increased, whereas branches did not differ significantly.
