      Pengaruh Penambahan Polisorbat pada Media Pengayaan terhadap Persentase Recovery Pengujian Mikrobiologi Produk Serbuk Effervescent di PT XYZ

      Abstract
      Uji mikrobiologi merupakan bagian penting dalam menjamin keamanan serta masa simpan produk makanan dan minuman. Namun, produk yang mengandung bahan antimikroba, seperti serbuk effervescent dari PT XYZ, dapat menghambat pertumbuhan mikroorganisme uji sehingga menurunkan validitas hasil pengujian. Untuk mengatasi masalah tersebut, penelitian ini bertujuan mengevaluasi efektivitas penambahan polisorbat (Tween 80) sebagai agen penetral dalam media pengayaan pada pengujian Angka Lempeng Total (ALT). Penelitian menggunakan tiga mikroorganisme uji, yaitu Bacillus subtilis, Staphylococcus aureus, dan Escherichia coli, yang diuji dalam media pengayaan dengan penambahan polisorbat pada konsentrasi 0,1 %, 0,5 %, dan 1 %. Hasil penelitian menunjukkan bahwa penambahan polisorbat meningkatkan keandalan uji mikrobiologi pada produk yang mengandung bahan antimikroba, seperti serbuk effervescent. Konsentrasi polisorbat 0,5 % memberikan hasil paling optimal dengan tingkat recovery mikroba = 70 % sesuai standar United States Pharmacopeia.
       
      Microbiological testing is an essential part of ensuring the safety and shelf life of food and beverage products. However, products containing antimicrobial agents, such as the effervescent powder from PT XYZ, can inhibit the growth of test microorganisms, thereby reducing the validity of the test results. To address this issue, this study aims to evaluate the effectiveness of adding polysorbate (Tween 80) as a neutralizing agent in the enrichment media for the Total Plate Count (TPC) method. The study used three test microorganisms, Bacillus subtilis, Staphylococcus aureus, and Escherichia coli, which were tested in enrichment media containing polysorbate at concentrations of 0,1 %, 0,5 %, and 1 %. The results showed that the addition of polysorbate improved the reliability of microbiological testing for products containing antimicrobial agents, such as effervescent powder. A polysorbate concentration of 0,5 % was found to be optimal, with all microorganisms achieving a recovery rate of = 70 %, in accordance with United States Pharmacopeia standards.
