      KARAKTERISTIK FISIKOKIMIA DAN AKTIVITAS ANTIDIABETES EKSTRAK KULIT KAYU MANGIUM DAN NANOFITOSOMNYA BERDASARKAN UJI IN VIVO

      Abstract
      Ekstrak etanol kulit kayu mangium (Acacia mangium Willd.) bagian luar (outerbark)/EE-KLKM hasil ekstraksi bertingkat menggunakan Ultrasound Assisted Extraction (UAE) berdasarkan uji in vitro memiliki aktivitas antidiabetes. Penelitian ini bertujuan menentukan rendemen EE-KLKM, pembuatan dan karakterisasi fisikokimia nanofitosomnya (NF-KLKM), serta menguji aktivitas antidiabetes EE-KLKM dan NF-KLKM secara in vivo. Ekstraksi bertingkat KLKM dengan pelarut n-heksana, etil asetat, dan etanol menggunakan metode UAE selama 45 menit. Nanofitosom KLKM (NF-KLKM) dibuat dengan nisbah EE-KLKM:lesitin kedelai sebesar 1:1 dan diuji karakteristiknya. Uji in vivo dilakukan pada mencit jantan selama 21 hari dengan dosis ekuivalen. Rendemen tertinggi diperoleh dari EE-KLKM sebesar 16,05±0,32%. NF-KLKM berukuran 378,18±8,14 nm dengan nilai polidispersitas 0,23±0,02. EE-KLKM dosis 125 mg/kgBB menurunkan gula darah hingga 250 mg/dL dan meningkatkan insulin hingga 4546,75 pg/mL, sedangkan NF-KLKM dosis ekuivalen 250 mg/kgBB menurunkan gula darah hingga 220 mg/dL dan meningkatkan insulin hingga 4371,25 pg/mL.
       
      The ethanol extract of mangium (Acacia mangium Willd.) outer bark (EE-KLKM), obtained through sequential extraction using Ultrasound-Assisted Extraction (UAE), has shown in vitro antidiabetic activity. This study aimed to determine the yield of EE-KLKM, prepare and characterize its nanophytosomes (NF-KLKM), and evaluate their in vivo antidiabetic activity. Sequential extraction of KLKM was performed with n-hexane, ethyl acetate, and ethanol for 45 minutes using UAE. NF-KLKM was prepared at an EE-KLKM:soybean lecithin ratio of 1:1 and characterized. In vivo tests were conducted on male mice for 21 days at equivalent doses. The highest yield was obtained from EE-KLKM at 16.05±0.32%. NF-KLKM had a particle size of 378.18±8.14 nm and a polydispersity index of 0.23±0.02. EE-KLKM at 125 mg/kg BW reduced blood glucose to 250 mg/dL and increased insulin to 4546.75 pg/mL, while NF-KLKM at the equivalent dose of 250 mg/kg BW reduced blood glucose to 220 mg/dL and increased insulin to 4371.25 pg/mL.
