      Potensi Pediococcus pentosaceus E7 dalam Mendegradasi Selulosa dan Pektin untuk Produksi Serat dari Limbah Daun Mahkota Nanas

      Abstract
      Daun mahkota nanas merupakan limbah pertanian yang banyak mengandung selulosa dan pektin, namun belum dimanfaatkan secara maksimal menjadi produk bernilai ekonomi. Penelitian ini bertujuan menganalisis kemampuan Pediococcus pentosaceus E7 untuk mendegradasi selulosa dan pektin pada daun mahkota nanas melalui fermentasi substrat padat. Penelitian dilakukan menggunakan Rancangan Acak Lengkap (RAL) faktorial dengan dua faktor. Faktor pertama adalah konsentrasi kultur bakteri terdiri atas tiga taraf yaitu tanpa penambahan kultur bakteri, penambahan kultur P. pentosaceus E7 (8,25 log CFU/mL) sebanyak 10%, dan 20%. Faktor kedua adalah durasi fermentasi terdiri atas tiga taraf yaitu 0,7, dan 14 hari. Hasil penelitian menunjukkan bahwa P. pentosaceus E7 memiliki kemampuan selulolitik dan pektinolitik ditandai dengan terbentuknya zona bening pada media CMC dan pektin padat. Perlakuan penambahan kultur bakteri 20% selama fermentasi 14 hari menghasilkan gula pereduksi, gula total, dan rendemen tertinggi secara berturut-turut sebesar 0,87 mg/mL, 1,03 mg/mL, dan 60,81% serta pH dan derajat polimerisasi terendah masing-masing sebesar 3,66 dan 1,03. Perlakuan tanpa penambahan kultur pada durasi fermentasi yang sama hanya menghasilkan gula pereduksi 0,26 mg/mL, total gula 0,51 mg/mL, rendemen 41,36%, pH 4,87, serta derajat polimerisasi 1,99. Hasil ini menunjukkan bahwa P. pentosaceus E7 efektif dalam meningkatkan degradasi selulosa dan pektin daun mahkota nanas.
       
      Pineapple crown leaves are agricultural waste rich in cellulose and pectin, but remain underutilized economically. This study analyzes the ability of Pediococcus pentosaceus E7 to degrade cellulose and pectin in pineapple crown leaves through solid substrate fermentation. The study was conducted using a completely randomized design (CRD) factorial with two factors. The first factor was bacterial culture concentration, consisting of three levels: no bacterial culture addition, addition of P. pentosaceus E7 culture (8.25 log CFU/mL) at 10%, and 20%. The second factor was fermentation duration, consisting of three levels: 0, 7, and 14 days. The results showed that P. pentosaceus E7 possesses cellulolytic and pectinolytic capabilities, indicated by the formation of clear zones on CMC and solid pectin media. The treatment with 20% bacterial culture during 14 days of fermentation produced the highest reducing sugar (0.87 mg/mL), total sugar (1.03 mg/mL), and yield (60.81%), along with the lowest pH (3.66) and degree of polymerization (1.03). The treatment without bacterial culture with same fermentation duration only produced reducing sugar (0.26 mg/mL), total sugar (0.51 mg/mL), yield (41.36%), pH (4.87), and polymerization degree (1.99). These results indicate that P. pentosaceus E7 is effective at degrading cellulose and pectin in pineapple crown leaves.
