      Aplikasi Nanopartikel Perak (Ag-NP) untuk Menekan Infeksi Tobacco mosaic virus Isolat Mentimun

      Abstract
      Tobacco mosaic virus (TMV) isolat mentimun merupakan virus yang berpotensi sebagai penyakit penting pada tanaman Cucurbitaceae di Indonesia. Upaya pengelolaan TMV masih terbatas. Teknologi nanopartikel berkembang pesat pemanfaatannya dalam berbagai bidang, tetapi pemanfaatannya dalam proteksi tanaman masih sedikit. Penelitian bertujuan menguji kemampuan nanopartikel perak (Ag-NP) untuk menekan infeksi TMV pada tanaman mentimun. Ag-NP disintesis menggunakan ekstrak daun bugenvil sebagai reduktor. Ag-NP diaplikasikan dengan penyemprotan daun sebelum dan setelah penularan TMV pada konsentrasi 50 ppm dan 60 ppm. Parameter penyakit dan agronomis diamati setiap minggu. Ag-NP pada konsentrasi 60 ppm sebelum inokulasi virus secara signifikan menurunkan insidensi dan keparahan penyakit, memperpanjang masa inkubasi, dan menurunkan titer virus, jika dibandingkan perlakuan Ag-NP setelah inokulasi virus. Selain itu, perlakuan Ag-NP mampu mempertahankan kadar klorofil dan menunjukkan pertumbuhan tanaman sebanding dengan kontrol sehat. Hasil penelitian menunjukkan bahwa Ag-NP tidak hanya memiliki efek antivirus, tetapi juga mampu memperkuat fisiologi tanaman. Implikasi hasil ini membuka peluang pengembangan Ag-NP berbasis hayati sebagai komponen dalam strategi pengendalian terpadu virus tanaman yang lebih berkelanjutan.
       
      The Tobacco mosaic virus (TMV) isolate of cucumber is a virus that has the potential to be an important disease of Cucurbitaceae plants in Indonesia. TMV management efforts are still limited. Nanoparticle technology is rapidly developing its utilisation in various fields, but its use in plant protection is still few. The study aimed to test the ability of silver nanoparticles (Ag-NP) to suppress TMV infection in cucumber plants. Ag-NPs were synthesised using bougainvillea leaf extract as reductant. Ag-NP was applied by foliar spraying prior and post TMV transmission at concentrations of 50 ppm and 60 ppm. The disease and agronomic parameters were observed weekly. Ag-NP at a concentration of 60 ppm before virus inoculation significantly reduced disease incidence and severity, prolonged the incubation period, and reduced virus titre in compared to Ag-NP treatment post virus innoculation. In addition, Ag-NP treatment prior TMV infection showed able to, maintained chlorophyll levels and plant growth comparable to healthy controls. These findings suggest that Ag-NPs have antiviral effect and ability to strengthen plant physiology. The implications of these results open opportunities for the development of bio-based Ag-NPs as a component in a more sustainable integrated control strategy for plant viruses.
