      Abstract
      MOHAMMAD NAJAFREIL DARRICK WARDHANA. Isolasi dan Karakterisasi Bakteri Heterotrof pada Suhu Inkubasi yang Berbeda dari Sedimen Laut Dalam di Perairan Banggai, Indonesia. Dibimbing oleh MEUTIA SAMIRA ISMET dan ENDANG SUNARWATI SRIMARIANA. Laut dalam mempunyai keanekaragaman hayati bakteri yang banyak namun cenderung lebih sulit untuk diisolasi. Kesulitan mengisolasi bakteri laut dalam dikarenakan bakteri laut dalam sensitif terhadap suhu. Sensitivitas terhadap suhu menjadi faktor utama dalam proses inkubasi bakteri laut dalam. Penelitian ini bertujuan mengisolasi dan karakterisasi bakteri heterotrof yang berasal dari sedimen laut dalam di Perairan Banggai, Indonesia, dan mengetahui perbandingan jumlah bakteri dari kondisi suhu yang berbeda, serta mengidentifikasi genus bakteri dari sampel sedimen laut dalam. Isolasi bakteri dari sedimen laut dalam menggunakan media Zobell 2216E dengan suhu 4?, 20?, dan 28? diperoleh 17 isolat. Bakteri heterotrof dapat digunakan dalam bidang bioremediasi dan bioteknologi. Dominansi Gram di sedimen laut dalam yaitu Gram negatif berbentuk basil karena dapat bertahan dalam kondisi lingkungan yang ekstrem. Identifikasi terhadap 17 isolat mendapat kecocokan dengan genus Enterobacter dan Psychrobacter.

Supervised by MEUTIA SAMIRA ISMET and ENDANG SUNARWATI SRIMARIANA. The deep-sea harbors a vast diversity of bacterial species however these microorganisms are generally more challenging to isolate. The difficulty in isolating deep-sea bacteria is due to their sensitivity to temperature. The difficulty in isolating deep-sea bacteria is due to their sensitivity to temperature. This research objective is to isolate and characterize heterotrophic bacteria from deep-sea sediments in the Banggai waters, Indonesia, determine the comparison of bacterial counts under different temperature conditions, and identify the bacterial genera from deep-sea sediment samples. Bacterial isolation from deep-sea sediments using Zobell 2216E medium at temperatures of 4?, 20?, and 28? collected 17 isolates. Heterotrophic bacteria can be utilized for bioremediation and biotechnology. Gram-negative bacillus shape dominated in deep-sea sediments because they can survive in extreme environmental conditions. Identification of the 17 isolates showed a match with the genera Enterobacter and Psychrobacter.
